Skin Necrosis

Skin necrosis is the irreversible death of skin cells and tissue, usually caused by severe disruption of blood supply, infection, trauma, or chemical injury. When oxygen and nutrient delivery falls below cellular requirements, ischemia damages tissue, disrupts membrane integrity, and can produce discoloration, loss of sensation, ulceration, and tissue breakdown. In medicine, recognizing skin necrosis helps clinicians identify conditions such as pressure injuries, vascular disease, burns, and severe soft-tissue infections, assess the extent of injury, and guide treatment. Evaluation may include physical examination, vascular assessment, imaging, and laboratory testing, while management focuses on restoring perfusion, controlling infection, removing nonviable tissue, and supporting wound healing.

Necrosis is considered as an “accidental” or unexpected form of cell death that ends in cell lysis. The first noticeable mention of “necrosis” was in 1859 when Rudolf Virchow used this term to describe advanced tissue breakdown in his compilation titled “Cell Pathology”. Morphological Manifestations of Necrosis Necrotic cells show different types of morphological appearance depending on the type of tissue and infection. In coagulative necrosis, cells become anucleated and die, but their...

Source: Chen, Y. et. al., Evaluating Virulence and Pathogenesis of Aeromonas Infection in a Caenorhabditis elegans Model. J. Vis. Exp. (2018)This video demonstrates a muscle necrosis assay in C. elegans (Caenorhabditis elegans) using pathogenic Aeromonas strains. By feeding green fluorescent protein (GFP)-expressing worms on Aeromonas-seeded plates, the assay models bacterial infection and reveals tissue damage through fluorescence imaging. The progressive loss of muscle structure and GFP...

Necrosis is a form of irreversible cell death caused by severe injury such as ischemia, toxins, or trauma. Unlike programmed cell death, it is an uncontrolled, pathological process that typically provokes inflammation in surrounding tissues.Pathophysiologic ChangesNecrosis begins when cells sustain critical damage, leading to swelling of organelles, particularly mitochondria, and rapid ATP depletion. As energy levels decline, membrane ion pumps fail, leading to calcium influx and eventually,...
