Subcutaneous Pouch

A subcutaneous pouch is a surgically created space beneath the skin that accommodates an implanted device, reservoir, tubing, or tissue while separating it from the external environment. In neuroscience research, investigators form the pouch by carefully separating skin from underlying connective tissue, place the selected component within it, and close the incision so the implant remains positioned while connections can be routed to a target site or accessed externally when required. This approach supports chronic neural recording, stimulation, and controlled drug delivery studies by improving device stability and protecting components during repeated measurements. Proper tissue handling and monitoring are essential for reliable results and animal welfare.

This video describes the procedure for generating an air pouch in the subcutaneous tissue of mice and collecting inflammatory exudates in response to lipopolysaccharide administration in vivo. This video also covers the analysis of the neutrophil infiltration in the air pouch.

Developing an Experimental Aneurysm Model in a Rabbit Using an Elastase-Treated Vessel Pouch

Source: Wanderer, S., et al. Arterial Pouch Microsurgical Bifurcation Aneurysm Model in the Rabbit. J. Vis. Exp. (2020)This video demonstrates the creation of an experimental aneurysm model in a rabbit using an elastase-treated vessel pouch. After excising a segment of the right CCA, the pouch is treated with elastase, then sutured to the left CCA to restore blood flow. The resulting stress on the weakened pouch walls induces aneurysm formation.

An optimized technique for the microsurgical creation of arterial bifurcation aneurysms mimicking bifurcation human cerebral aneurysms is described. A venous pouch is sutured into an artificially created true bifurcation of both common carotid arteries. Facilitated microsurgical techniques and aggressive postoperative anticoagulation and analgesia lead to minimized morbidity rates and high aneurysm patency rates.

This protocol describes a unique experimental model of implant-related infections that enables the simultaneous incubation of two implants with bacteria under identical conditions within a single mouse. It also allows precise assessment of biofilm formation on implant surfaces using optimized comparative analytical methods, demonstrating advanced techniques for evaluating biomaterials' antimicrobial properties.
