Compound Screening Cascade

A compound screening cascade is a staged strategy for evaluating chemical compounds, designed to identify promising drug candidates while efficiently eliminating inactive, nonspecific, or toxic molecules. Compounds first undergo a broad, often high-throughput primary assay, and active hits then advance through progressively more selective secondary assays that assess potency, mechanism of action, selectivity, cellular activity, and early safety properties. In pharmacology, this process connects molecular screening with biological validation, helping researchers prioritize compounds for lead optimization and further preclinical development. By combining complementary tests, a screening cascade reduces resources spent on weak candidates and improves confidence that retained compounds act through relevant therapeutic pathways.

Rab GTPases act in a regulated cascade during membrane fusion, helping the lipid bilayers mix. The Rab family of proteins are active when bound to GTP, and inactive when bound to GDP. Hence, they act as guanine nucleotide-dependent molecular switches. Rab-GTP recognizes and binds to long or short-range tethering proteins to capture the target vesicle. These tethers coordinate with SNAREs on the vesicle and the target membrane to assemble the trans SNARE complex that locks the mixing bilayers.
