Rosiglitazone

Rosiglitazone is an oral thiazolidinedione antidiabetic drug that improves insulin sensitivity in people with type 2 diabetes. It activates peroxisome proliferator-activated receptor gamma (PPARγ), a nuclear receptor that alters gene transcription involved in glucose and lipid metabolism, thereby increasing insulin-responsive glucose uptake and reducing insulin resistance. Its pharmacological effects can lower blood glucose without directly stimulating pancreatic insulin release, but they may also cause fluid retention, weight gain, and an increased risk of heart failure in susceptible patients. Rosiglitazone therefore serves as a model for studying nuclear receptor pharmacology, metabolic regulation, drug safety, and individualized diabetes treatment.
