In TLC, the stationary phase is a thin layer of polar adsorbent material, typically silica gel, fixed to a glass or plastic plate. The mobile phase is a non-polar liquid that moves up the plate by capillary action, carrying sample components with it. Compounds separate based on their interactions with both phases.

Less polar compounds spend more time in the mobile phase and travel farther up the plate. More polar compounds are attracted to the polar stationary phase and do not migrate as far. This differential movement based on polarity allows the separation of mixture components.

An appropriate mobile phase should be non-polar and produce well-separated rings, with the outermost ring about 50% of the distance to the solvent front. If the mobile phase is too non-polar, the sample remains near the baseline. If too polar, all sample migrates with the solvent front. Two miscible solvents can be mixed to achieve desired properties.

Keeping the spot small leads to better separation and resolution of compounds on the plate. If more sample is needed, multiple applications can be made at the same location, allowing the solvent to dry between applications. This prevents disturbing the stationary phase and ensures optimal separation results.

The retardation factor, or Rf, is the ratio of the distance a compound travels to the distance the mobile phase travels. It is calculated by measuring the distance from the baseline to the spot and dividing by the distance from the baseline to the solvent front. Rf values depend on solvent choice, adsorbent thickness, temperature, and sample size.

Colorless compounds can be visualized using a UV lamp set to short wave, which causes the compound to block the background fluorescence of the plate. Alternatively, potassium permanganate, an oxidizing agent, can be used as a stain. The plate is dipped in the stain and heated in a fume hood to reveal spots.

TLC can monitor chemical reaction progression by tracking starting material and product bands over time. It can separate and isolate specific compounds from natural sources for further analysis. TLC can also be used in bioassays to identify compounds with antimicrobial properties by placing separated bands on bacterial cultures.
