Replication in Eukaryotes

Die meisten prokaryotischen Faktoren, die bei der Replikation verwendet werden, haben &#xC4;quivalente, die bei der eukaryotischen DNA-Duplizierung &#xE4;hnlich wirken.
 Dieser Prozess beginnt an einem Replikationsursprung, an den sich ein Erkennungskomplex bindet.
 Helicase wird dann von der Stelle angezogen und trennt die DNA-Str&#xE4;nge, wodurch eine Blase mit zwei Gabeln entsteht.
 Primase trifft auch ein und erzeugt RNA-Primer, die bei Bewegung der Helikase die DNA-Polymerase mit einer neuen DNA verl&#xE4;ngern.
 Wie bei den Prokaryonten w&#xE4;chst der neu gebildete Leitstrang kontinuierlich, entlang der Replikationsgabel, umgekehrt wird der Leitstrang in kleinen Okazaki-Fragmenten hergestellt, die gegen&#xFC;ber der Gabel verlaufen.
 Aufgrund mehrerer Faktoren erzeugt die DNA-Vorlage, mit der der f&#xFC;hrende Strang in der einen H&#xE4;lfte dieser Struktur erzeugt wird, den isolierenden Strang in der anderen.
 Dabei gibt es verschiedene Replikationsurspr&#xFC;nge in einem linearen eukaryontischen Chromosom und die Replikation endet, wenn die zugeh&#xF6;rigen Zellen zusammenwachsen.
 Die Primer werden dann &#xFC;ber Enzyme, wie z.
 B.
 RNAse, eliminiert und zur DNA hin aufgel&#xF6;st.
 Danach bindet die DNA-Ligase alle Segmente aneinander, aber wenn der Endprimer aus dem isolierenden Strang verschwindet, bleibt der Platz frei und es gibt einen unkopierten Abschnitt der DNA-Vorlage, der an ihn st&#xF6;&#xDF;t.
 Um dies zu bek&#xE4;mpfen, bindet sich ein Enzym namens Telomerase an den &#xFC;berstehenden Bereich und verl&#xE4;ngert ihn mit einer nicht kodierenden DNA-Sequenz.
 Primase und DNA-Polymerase wirken auf diesen und bilden eine Telomerkappe, die vor Verlust der DNA-Kodierung aus dem Isolierstrang bei mehrfachen Replikationen sch&#xFC;tzt.
 Die eukaryontische DNA-Replikation endet mit 2 DNA-Molek&#xFC;len mit je einem elterlichen und neu synthetisierten Strang, zahlreichen Replikationsurspr&#xFC;ngen und Telomeren.

 In eukaryotischen Zellen ist die DNA-Replikation hoch konserviert und wird streng reguliert. Mehrere lineare Chromosomen m&#xFC;ssen vor der Zellteilung mit hoher Genauigkeit vervielf&#xE4;ltigt werden. Daher gibt es viele Proteine, die spezielle Rollen im Replikationsprozess ausf&#xFC;llen. Die Replikation erfolgt in drei Phasen: Initiation, Elongation und Termination. Sie resultiert in zwei vollst&#xE4;ndigen Chromosomens&#xE4;tzen im Zellkern.
 
 <h4>Viele Proteine koordinieren die Replikation am Replikationsursprung</h4>
 
 Die eukaryotische Replikation folgt vielen Prinzipien der prokaryotischen DNA-Replikation. Da das Genom jedoch viel gr&#xF6;&#xDF;er ist und die Chromosomen eher linear als zirkul&#xE4;r sind, ben&#xF6;tigt der Prozess mehr Proteine und hat somit einige wesentliche Unterschiede. Die Replikation erfolgt gleichzeitig an mehreren Replikationsurspr&#xFC;ngen entlang jedes Chromosoms. Initiatorproteine erkennen und binden an den Replikationsursprung und aktivieren die Helikase, um die DNA-Doppelhelix zu entwickeln. An jedem Replikationsursprung bilden sich zwei Replikationsgabeln. Die Primase f&#xFC;gt dann kurze RNA-Primer an die einzelnen DNA-Str&#xE4;nge an, die als Ausgangspunkt f&#xFC;r die Bindung der DNA-Polymerase dienen, welche dann beginnt die Sequenz zu kopieren. Die DNA kann nur in der 5 bis 3 Richtung synthetisiert werden, so dass die Replikation beider Str&#xE4;nge von einer einzigen Replikationsgabel aus in zwei verschiedene Richtungen verl&#xE4;uft. Der Leitstrang wird kontinuierlich synthetisiert, w&#xE4;hrend der Folgestrang in kurzen Abschnitten mit 100-200 Basenpaaren, den sogenannten Okazaki-Fragmenten, synthetisiert wird. Sobald der Gro&#xDF;teil der Replikation abgeschlossen ist, entfernen RNase-Enzyme die RNA-Primer und die DNA-Ligase verbindet jegliche L&#xFC;cken in dem neuen Strang.
 
 <h4>Verteilung der Replikationsarbeit unter den Polymerasen</h4>
 
 Die Arbeitsbelastung beim Kopieren von DNA in Eukaryoten wird auf mehrere verschiedene Typen der DNA-Polymerase-Enzyme verteilt. Die wichtigsten Familien der DNA-Polymerasen in allen Organismen werden nach der &#xC4;hnlichkeit ihrer Proteinstrukturen und Aminos&#xE4;uresequenzen kategorisiert. Die ersten Familien, die entdeckt wurden, werden als A, B, C und X bezeichnet, die Familien Y und D wurden sp&#xE4;ter identifiziert. Zu den Polymerasen der Familie B in Eukaryoten geh&#xF6;ren Pol &#x3B1;, die auch als Primase an der Replikationsgabel fungiert, und Pol &#x3B4; und &#x3B5; die Enzyme, die die meiste Arbeit der DNA-Replikation auf dem Leit- und dem Folgestrang der Matrize verrichten. Andere DNA-Polymerasen sind f&#xFC;r Aufgaben wie die Reparatur von DNA-Sch&#xE4;den, das Kopieren von mitochondrialer und Chloroplasten-DNA und das Auff&#xFC;llen von L&#xFC;cken in der DNA-Sequenz auf dem Folgestrang nach Entfernung der RNA-Primer verantwortlich.
 
 <h4>Telomere sch&#xFC;tzen die Enden der Chromosomen vor dem Abbau</h4>
 
 Da die eukaryotischen Chromosomen linear sind, sind ihre Enden f&#xFC;r einen Abbau anf&#xE4;llig. Um wichtige genetische Informationen vor Sch&#xE4;den zu sch&#xFC;tzen, enthalten die Enden der Chromosomen viele nicht-kodierende Wiederholungen an hochkonservierter G-reicher DNA: die Telomere. Ein kurzer einzelstr&#xE4;ngiger 3-&#xDC;berhang an jedem Ende des Chromosoms interagiert mit spezialisierten Proteinen, die das Chromosom im Zellkern stabilisieren. Aufgrund der Art und Weise, wie der Folgestrang synthetisiert wird, kann ein kleiner Teil der Telomersequenzen nicht mit jeder Zellteilung repliziert werden. Dadurch verk&#xFC;rzen sich die Telomere im Laufe vieler Zellzyklen allm&#xE4;hlich und k&#xF6;nnen als Marker der Zellalterung gemessen werden. Bestimmte Zellpopulationen, wie Keimzellen und Stammzellen, exprimieren Telomerase, ein Enzym, das die Telomere verl&#xE4;ngert, so dass die Zelle mehr Zellzyklen durchlaufen kann, bevor sich die Telomere verk&#xFC;rzen.

Eukaryotic DNA copying is tightly regulated and must duplicate many linear chromosomes before cell division. The process is highly conserved and uses many proteins with specialized roles. It moves through initiation, elongation, and termination, and it ends with two complete sets of chromosomes in the nucleus.

Eukaryotic replication shares basic steps with prokaryotic DNA replication, but the larger genome and linear chromosomes require more proteins. Replication begins at multiple origins of replication along each chromosome. Initiator proteins bind the origin and recruit helicase, which unwinds the DNA double helix. Two replication forks form at each origin. Primase then adds short RNA primers to the single strands, and these primers let DNA polymerase bind and start copying the DNA.

DNA synthesis can only proceed in the 5′ to 3′ direction, so the two strands are copied in different ways at each replication fork. The leading strand is made continuously. The lagging strand is made in short stretches of 100 to 200 base pairs called Okazaki fragments. After most replication is finished, RNase enzymes remove the RNA primers, and DNA ligase seals the remaining gaps in the new strand.

Eukaryotic cells use several DNA polymerases to divide the work of replication. Polymerase families are grouped by similarities in protein structure and amino acid sequence. Early families were called A, B, C, and X, and Y and D were identified later. In eukaryotes, family B polymerases include Pol alpha, which also works as a primase at the replication fork, and Pol delta and epsilon, which do most of the DNA copying on the leading and lagging strands. Other polymerases help repair DNA damage, copy mitochondrial and plastid DNA, and fill gaps left on the lagging strand after RNA primers are removed.

Because eukaryotic chromosomes are linear, their ends need special protection. These ends contain telomeres, which are long non-coding repeats of G-rich DNA. A short single-stranded 3′ overhang at each end interacts with specialized proteins that help stabilize the chromosome in the nucleus. Still, the lagging strand cannot fully copy the telomeric DNA during every cell division, so telomeres become shorter over many cell cycles and can serve as a marker of cellular aging. Some cells, including germ cells and stem cells, express telomerase, an enzyme that lengthens telomeres and allows more rounds of cell division before shortening occurs.

EXPLANATION: Discussions of DNA in earlier grades have been limited to discussions of genes and the role of genes in the appearance and activities of an organism. Biology 1 represents a student's introduction to a molecular model of a DNA as well as the organization of DNA into genes and genes subsequently into chromosomes. Students should address interactions between genes in proteins which regulate both the shape and reproduction of DNA molecules. It is important to note that not all segments of DNA are transcribed for proteins. Portions of DNA are involved in regulating the expression of genes or affect the structure of the chromosomes (replication origin, centromeres, and telomeres).
