Mutations

A mutation is a change in the DNA sequence, which arises due to errors during DNA replication or from environmental factors such as radiation or certain chemicals. Since DNA stores instructions for making proteins, a change in DNA can alter the amino acid sequence&#x2014;the order of amino acids in a protein&#x2014;which may affect its structure and function.One common type of mutation is a point mutation, where a single nucleotide changes. These include silent, missense, and nonsense mutations.Silent mutations, as the name suggests, do not change the amino acid sequence of a protein.For example, if the codon CCA changes to CCG, it still codes for the amino acid proline. Because the amino acid sequence stays the same, the protein&#x2019;s structure remains unchanged, so it retains its normal function, such as converting a substrate into a product.Missense mutations happen when a change in the DNA sequence replaces one amino acid with another. For example, arginine may replace proline. This change can affect the protein&#x27;s shape or function.Nonsense mutations happen when a codon for an amino acid changes into a stop codon. This signals the cell to stop translation early and produces a truncated protein that is often nonfunctional.Another type of DNA mutation is a frameshift mutation. It happens when one or more nucleotides are inserted or deleted.Because codons are read in groups of three, this shift changes every codon that follows. As a result, a series of new codons forms that code for different amino acids and leads to an abnormal protein.Depending on how they alter protein structure and function, mutations can be neutral, causing no change in function; harmful, producing nonfunctional or truncated proteins; or beneficial, enhancing protein activity.

Mutations are changes in the sequence of DNA. These changes can occur spontaneously during DNA replication or be induced by environmental factors. Mutations can be characterized in several ways: by whether and how they alter the amino acid sequence of the protein, by the scale of the DNA affected, and by whether they affect somatic or germline cells.Consequences of Point Mutations at the Molecular Level Mutations that affect a single nucleotide are called point mutations. When point mutations arise within genes, the consequences can vary in severity depending on how they alter the encoded amino acid sequence. A silent mutation does not change the amino acid sequence and is often neutral. A missense mutation changes a single amino acid, and the effects might be serious if the change alters the function of the protein. A nonsense mutation introduces a stop codon, truncating the protein and likely rendering it nonfunctional. Insertions or deletions of nucleotides can cause frameshift mutations if they alter the reading frame, affecting all downstream codons and often severely disrupting protein function.Chromosomal Alterations Are Large-Scale Mutations The most drastic type of mutation, chromosomal alteration, changes the structure or number of chromosomes. Structural changes can include deletions, duplications, inversions, and translocations of large DNA segments within a single chromosome. Numerical abnormalities such as aneuploidy involve the gain or loss of entire chromosomes. Chromosomal mutations are typically far more serious than point mutations because they affect many genes and regulatory elements. Chromosomal alterations can be detected by karyotyping the affected cell.Only Germline Mutations Are Inherited Mutations can arise in any cell, but only germline mutations&#x2014;those present in egg and sperm cells&#x2014;can be transmitted to offspring. Hereditary diseases are a subtype of genetic disorders caused by deleterious germline mutations. They can be autosomal, involving chromosomes 1 through 22, or sex-linked, involving the X or Y chromosome. One example of a hereditary disease is cystic fibrosis (CF), which primarily affects the lungs. It is caused by a deletion in the CFTR gene that removes a single amino acid from the CFTR protein. CF is an autosomal recessive disease, meaning that a person with one mutated copy of the gene and one normal copy will not develop the disease. On the other hand disease like Huntington&#x2019;s disease, a neurodegenerative disorder, are autosomal dominant, meaning that only one mutated copy of the gene is necessary for the disease to develop.Some Mutations Are Caused by Environmental Factors Both somatic mutations and germline mutations can arise spontaneously, but they can also be induced by environmental agents known as mutagens. These include environmental radiation or chemicals that damage DNA. One well-characterized environmental mutagen is ultraviolet (UV) radiation. UV radiation induces the formation of covalent bonds between adjacent thymine bases on the same DNA strand, creating thymine dimers that distort the DNA structure. The sun is a natural source of UV radiation. The most harmful UV-C rays are absorbed by the atmosphere, while UV-A and UV-B rays reach the Earth&#x27;s surface. Artificial sources of UV exposure include tanning beds, which transmit primarily UV-A rays with smaller amounts of UV-B.Fortunately, cells have mechanisms to repair damaged DNA, but sometimes the damage is not repaired before cell division, particularly in rapidly dividing cells such as skin cells, and mutations can accumulate. When these mutations affect genes that regulate cell growth and division, they can contribute to the development of cancer.

Mutations are changes in the DNA sequence. They can happen by chance during DNA copying, or they can be caused by environmental factors such as radiation and chemicals. Scientists also describe mutations by how much DNA they affect, how they change a protein, and whether they happen in body cells or germline cells.

Some mutations affect only one nucleotide, which is a point mutation. When a point mutation falls inside a gene, the outcome depends on how the amino acid sequence changes. A silent mutation leaves the amino acid the same and usually has no effect. A missense mutation changes one amino acid, and the result can be serious if the protein no longer works well.

Other point mutations can stop protein production early. A nonsense mutation creates a stop codon, which truncates the protein and often makes it nonfunctional. A frameshift mutation happens when nucleotides are inserted or deleted in protein-coding DNA. This shifts the reading frame and changes all downstream codons.

Some mutations are large-scale chromosomal alterations. These changes affect the physical structure of a chromosome and can include deletion, duplication, inversion, or the addition of DNA from another chromosome. Because they can involve many genes and regulatory elements, they are usually more serious than point mutations. Cells with these changes can be identified by karyotyping.

Only germline mutations are inherited. These mutations are present in egg and sperm cells and can pass to offspring, while somatic mutations stay outside the germline. Hereditary diseases are caused by harmful germline mutations. They can be autosomal, on chromosomes 1 through 22, or sex-linked, on the X or Y chromosome.

Cystic fibrosis is one example of a hereditary disease. It affects the lungs and is caused by a deletion in the CFTR gene that removes a single amino acid from the CFTR protein. It is autosomal recessive, so one normal copy can prevent disease. Huntington's disease is another example, and it is autosomal dominant, which means one mutated copy is enough for the disorder to develop.

Mutations can also come from mutagens, which are external agents that damage DNA. Ultraviolet, or UV, radiation is a well-known mutagen. It has more energy than visible light and can create thymine dimers when bases on the same DNA strand bond with each other. The sun is a natural source of UV radiation, and tanning beds are artificial sources that mainly emit UV-A with some UV-B.

Cells can repair DNA damage, but repair is not always complete before cell division. This matters most in rapidly dividing cells such as skin cells. If the damage affects genes that help control cell growth and division, it can contribute to cancer.
