The Central Dogma

Das zentrale Dogma der Molekularbiologie besagt, dass in DNA kodierte Informationen auf RNA übertragen werden, die dann die Synthese von Proteinen basierend auf diesen Anweisungen steuert.
 Zunächst wird bei der Transkription DNA als Matrize zur Synthese von Messenger-RNA (mRNA) verwendet, die eine Kopie des kodierenden Strangs darstellt, mit der Ausnahme, dass die Thymidine durch Uracils ersetzt werden.
 Als nächstes wandert die mRNA bei der Translation in Eukaryoten zu einem Ribosom.
 Hierbei binden Codone, Gruppen von drei Nucleotiden in der mRNA an komplementäre Sequenzen auf der Transfer-RNA, tRNA-Moleküle, von denen jedes abhängig von dem spezifischen Codon an eine bestimmte Aminosäure gebunden ist.
 Beispielsweise bindet das Codon CCA an eine an Prolin gebundene tRNA, während AGC an eine an Serin gebundene tRNA bindet.
 Auf diese Weise gibt der genetische Code die Reihenfolge an, in der die Aminosäuren im entstehenden Polypeptid angeordnet sind.
 Polypeptide werden dann oft zu funktionellen Proteinen weiterverarbeitet.

<h4>Überblick</h4>
 
 Die zentrale Lehre der Biologie besagt, dass die in der DNA geschriebene Information auf die Boten-RNA (mRNA) übertragen wird. Diese steuert anschließend die Proteinsynthese. Die Gesamtheit der Anweisungen, die es ermöglichen, die mRNA-Sequenz in Aminosäuren umzuschreiben, wird als genetischer Code bezeichnet. Die Allgemeingültigkeit dieses genetischen Codes hat den Fortschritt in der wissenschaftlichen Forschung, der Landwirtschaft und der Medizin vorangetrieben.
 
 Die RNA ist die fehlende Verbindung zwischen DNA und Proteinen</h4>
 
 Anfang 1900 entdeckten Wissenschaftler, dass die DNA alle Informationen speichert, die für zelluläre Funktionen benötigt werden und das Proteine die meisten Funktionen ausführen. Die Mechanismen der Umwandlung der genetischen Information in funktionelle Proteine blieben jedoch viele Jahre lang ungeklärt. Zunächst glaubte man, dass ein einzelnes Gen direkt in sein kodiertes Protein umgewandelt wird. Zwei entscheidende Entdeckungen in eukaryotischen Zellen stellten diese Theorie jedoch in Frage. Erstens findet die Proteinproduktion nicht im Zellkern statt. Zweitens ist DNA nicht außerhalb des Zellkerns vorhanden. Diese Erkenntnisse lösten die Suche nach einem Zwischenmolekül aus, welches die DNA mit der Proteinproduktion verbindet. Dieses Zwischenmolekül, das sich sowohl im Zellkern als auch im Zytoplasma befindet und mit der Proteinproduktion in Verbindung steht, ist die RNA.
 
 Während der Transkription wird RNA im Zellkern synthetisiert, wobei die DNA als Vorlage dient. Die neu synthetisierte RNA ist in der Sequenz ähnlich wie der DNA-Strang, außer dass Thymidin in der DNA durch Uracil in der RNA ersetzt wird. In Eukaryoten wird dieses primäre Transkript weiter prozessiert, wobei die nicht kodierenden Bereiche des Proteins entfernt werden. Zusätzlich wird den 3'- und 5'-Enden ein 3' poly-A Schwanz hinzugefügt. Diese gebildete mRNA wird dann ins Zytoplasma übertragen.
 
 <h4>Die Regeln für die Umsetzung der mRNA-Sequenz bilden den genetischen Code</h4>
 
 Die Translation erfolgt durch die Ribosome im Zytoplasma, wobei die in der mRNA kodierte Information in eine Aminosäurekette übersetzt wird. Ein Satz von drei Nukleotiden kodiert für eine Aminosäure und diese Tripletts werden als Codons bezeichnet. Das Regelwerk, das festlegt, welche Codons eine bestimmte Aminosäure produzieren, bildet den genetischen Code.
 
 <h4>Der genetische Code ist degeneriert</h4>
 
 Proteine werden in Eukaryoten aus 20 Aminosäuren gebildet. Die Kombination von vier Nukleotiden in Dreiergruppen liefert 64 (43) mögliche Codons. Das bedeutet, dass es möglich ist, dass einzelne Aminosäuren von mehr als einem Codon kodiert werden können. Der genetische Code wird daher als degeneriert bezeichnet. In vielen Fällen, aber nicht immer, unterscheiden sich Codons, die gleiche Aminosäuren kodieren nur im dritten Nukleotid des Tripletts. Die Codons GUU, GUC, GUA und GUG stehen beispielsweise alle für die Aminosäure Valin. AUG ist jedoch das einzige Codon, das die Aminosäure Methionin repräsentiert. Das Codon AUG ist auch das Codon, an dem die Proteinsynthese beginnt und wird daher Startcodon genannt. Durch diese Degeneration des Systems werden die schädlichen Auswirkungen von Mutationen minimiert. Eine Mutation (d.h. Austausch) an der dritten Position des Codons muss nicht unbedingt zu einer anderen Aminosäure führen.
 
 <h4>Die Universalität des genetischen Codes</h4>
 
 Mit wenigen Ausnahmen verwenden die meisten prokaryotischen und eukaryotischen Organismen den gleichen genetischen Code für die Proteinsynthese. Diese Universalität des genetischen Codes hat Fortschritte in der wissenschaftlichen Forschung, der Landwirtschaft und der Medizin ermöglicht. Beispielsweise kann menschliches Insulin heute in großem Maßstab in Bakterien hergestellt werden. Dies geschieht mit Hilfe der rekombinanten DNA-Technologie. Rekombinante DNA besteht aus genetischem Material verschiedener Spezies. Gene, die menschliches Insulin kodieren, werden mit bakterieller DNA verbunden und in eine Bakterienzelle eingefügt. Die Bakterienzelle führt dann die Transkription und Translation durch, um das in der rekombinanten DNA kodierte menschliche Insulin zu produzieren. Das daraus resultierende Insulin wird zur Behandlung von Diabetes eingesetzt.

The central dogma explains how genetic information moves from DNA to RNA to protein. This flow is a key idea in molecular biology. It helps show how cells use genes to make the molecules that carry out work.

DNA stores the instructions for making proteins. During transcription, a gene in DNA is copied into RNA. The RNA then carries that message to the next stage.

In translation, the RNA message is used to build a protein. Proteins do much of the cell’s work, so this process connects genes to traits and cell function. Understanding the central dogma helps students see how information in DNA becomes an active product in the cell.
