The DNA Replication Fork

In der Zelle beginnt die DNA-Replikation immer an bestimmten Stellen auf der DNA, die als Ursprung der Replikation bezeichnet werden.
Zu Beginn der Replikation bindet das Enzym DNA-Helikase an den DNA-Strang und bewegt sich entlang der DNA, wodurch sich die DNA abwickelt und trennt. 
Die resultierende Y-förmige Struktur, die aus den einzelnen DNA-Strängen gebildet wird, sieht aus wie eine zweizinkige Gabel und wird zu einem Ort der DNA-Replikation; daher ist diese Struktur als "Replikationsgabel" bekannt.
Diese separaten, einzelsträngigen DNA-Moleküle neigen dazu, doppelsträngige Haarnadelschleifen zu bilden oder sich mit dem anderen Strang zurückzuspulen. 
Um dies zu verhindern, binden einzelsträngige DNA-bindende Proteine (SSBs) an einzelsträngige DNA und hemmen das Zurückspulen.
Nun können die freigelegten Einzelstränge der DNA als Vorlagen für die Synthese der komplementären Tochterstränge dienen.

Das Genom eines Organismus muss effizient und fehlerfrei repliziert werden, um Wachstum und Überleben zu sichern. Die Replikationsgabel ist eine Y-förmige aktive Region, in der zwei DNA-Stränge getrennt und kontinuierlich repliziert werden. Die Kopplung von DNA-Entzippen und komplementärer Strangsynthese ist ein charakteristisches Merkmal einer Replikationsgabel. Organismen mit kleiner zirkulärer DNA, wie z.B. E. coli, haben oft nur einen einzigen Replikationsursprung; daher haben sie nur zwei Replikationsgabeln, eine in jede Richtung, die vom Ort der ursprünglichen Öffnung wegführt. Bei Organismen mit großen Genomen erfolgt die Replikation der DNA nicht von einem einzelnen Ausgangspunkt aus, sondern an vielen verschiedenen, lokalisierten Replikationsgabeln.

Das ungehinderte Fortschreiten der Replikationsgabel ist notwendig für die vollständige DNA-Replikation und die Stabilität des Genoms; jedoch wird die Replikationsgabel oft durch interne oder externe Faktoren aufgehalten, die ihr Fortschreiten verlangsamen oder stoppen können, was zu Replikationsstress führt. Replikationsstress verursacht genomische Instabilität, ein Kennzeichen von Krankheiten wie Krebs. Genomische Instabilität ist gekennzeichnet durch genomische Veränderungen und eine erhöhte Häufigkeit schädlicher Mutationen. Die Bewegung der Replikationsgabel kann aus mehreren Gründen stoppen. Zum Beispiel kann das Medikament Hydroxyurea den Pool an verfügbaren Nukleotiden für die Einbindung in den Tochterstrang erschöpfen und die Replikationsgabel zum Stillstand bringen. Weitere Probleme, die das Fortschreiten der Replikationsgabel behindern können, sind DNA-Läsionen, eine Kollision zwischen einer Replikationsgabel und einem Transkriptionskomplex sowie Defekte in den an der DNA-Replikation beteiligten Enzymen.

Die Zelle verfügt über eine Vielzahl von Reparaturmechanismen, um die gestoppte Replikationsgabel wieder in Gang zu setzen. S-Phasen-Checkpoints lassen keine Zellteilung zu, bis die DNA-Reparatur abgeschlossen ist. Zusätzlich kann eine erneute Priming der Gabel die DNA-Synthese durch Umgehung einer DNA-Läsion oder Blockade neu starten. Trotz dieser robusten Mechanismen können manchmal gestoppte Gabeln nicht wieder initiiert werden, was zum Zusammenbruch einer Gabel führt und die DNA-Replikation stoppt.

DNA replication stress and fork stalling can disrupt the copying of a cell’s genome. The replication fork is the Y-shaped site where DNA strands separate and new complementary strands are made at the same time. This linked unwinding and synthesis helps cells duplicate DNA efficiently and with fewer errors.

In organisms with small circular DNA, such as E. coli, replication often begins at a single origin. That setup creates two replication forks that move in opposite directions from the opening site. In organisms with large genomes, DNA copying starts at many local replication forks instead of one origin.

Smooth fork movement is needed for complete DNA replication and genome stability. When a fork slows or stops, the cell experiences replication stress. This stress can lead to genomic instability, which means more harmful mutations and other DNA changes. Genomic instability is also a hallmark of diseases such as cancer.

Several problems can stall a replication fork. Hydroxyurea is one example. It reduces the pool of nucleotides needed to build the daughter strand. DNA lesions, collisions between a replication fork and a transcription complex, and defects in DNA replication enzymes can also block fork progress.

Cells have repair systems that can restart stalled forks. S-phase checkpoints prevent cells from entering mitosis until DNA repair is finished. Fork repriming can also restart DNA synthesis by skipping over a DNA lesion or other block. Even so, some stalled forks cannot be restarted, and fork collapse stops DNA replication.
