 Organismen sind in der Lage, Nukleotid-Fehlpaarungen, die w&#xE4;hrend der DNA-Replikation auftreten, zu erkennen und zu reparieren. Dieser anspruchsvolle Prozess erfordert die Identifizierung des neuen Stranges und den Austausch der fehlerhaften Basen mit korrekten Nukleotide. Die Korrektur der Fehlpaarungen wird von mehreren Proteinen sowohl in Prokaryoten als auch in Eukaryoten koordiniert.
 
 <h4>Die MMR-Proteinfamilie spielt eine Schl&#xFC;sselrolle in der DNA-Mismatch-Reparatur</h4>
 
 Das menschliche Genom hat mehr als 3 Milliarden Basenpaare DNA pro Zelle. Vor der Zellteilung muss diese riesige Menge an genetischer Information vervielf&#xE4;ltigt werden. Trotz der Korrekturf&#xE4;higkeit der DNA-Polymerase tritt etwa alle 1 Million Basenpaare ein Kopierfehler auf. Eine Art von Fehler ist die Fehlpaarung der Nukleotiden, zum Beispiel die Paarung von A mit G oder T mit C. Solche Fehlpaarungen werden von der MMR-Proteinfamilie erkannt und repariert. Diese Proteine wurden zuerst in den Bakterien Escherichia coli (E. coli) genauer beschrieben. &#xC4;hnliche Arten kommen aber in allen Prokaryoten und Eukaryoten vor.
 
 Der Mutator S (MutS) leitet die Mismatch-Reparatur (MMR) ein, indem er die Fehlpaarung identifiziert und an sie bindet. Anschlie&#xDF;end identifiziert MutL, welcher Strang die neue Kopie ist. Nur der neue Strang muss repariert werden, w&#xE4;hrend der Matrizenstrang intakt bleiben muss. Wie kann die molekulare Maschinerie den neu synthetisierten DNA-Strang identifizieren?
 
 <h4>Neu synthetisierte DNA-Str&#xE4;nge unterscheiden sich von ihrem Matrizenstrang</h4>
 
 In vielen Organismen erhalten Cytosin-und Adeninbasen des neuen Stranges einige Zeit nach der Replikation eine Methylgruppe. Deshalb identifizieren sogenannte Mut-Proteine neue Str&#xE4;nge. Sie erkennen Sequenzen, die noch nicht methyliert sind. Au&#xDF;erdem ist es wahrscheinlicher, dass der neu synthetisierte Strang in Eukaryoten kleine Br&#xFC;che, auch Strangbr&#xFC;che genannt, aufweist. Die MMR-Proteine k&#xF6;nnen so den fehlerhaften Strang identifizieren und ihn gezielt reparieren.
 
 Nach der Identifizierung des neuen Stranges schneiden die Nukleaseenzyme die betroffene Region und schneiden die falschen Nukleotide aus. Als n&#xE4;chstes f&#xFC;gt die DNA-Polymerase die richtigen Nukleotide ein. Die DNA-Ligase verkn&#xFC;pft das Zucker-Phosphat-R&#xFC;ckgrat der DNA, wodurch der Mismatch-Reparaturprozess abgeschlossen wird.
 
 <h4>Defekte im Mismatch-Reparaturmechanismus k&#xF6;nnen Krebs ausl&#xF6;sen</h4>
 
 Das menschliche Homolog von MutS wird als Mutator S-Homolog 2 (MSH2) bezeichnet. Wenn die Funktion von MSH2 beeintr&#xE4;chtigt wird, werden Punktmutationen und Frameshift-Mutationen im gesamten Genom nicht mehr richtig repariert. Folglich haben Menschen, die eine einzige Kopie eines solchen beeintr&#xE4;chtigten MSH2 tragen, eine h&#xF6;here Wahrscheinlichkeit, an Krebs zu erkranken.
 
 <h4>Mutationen, die nicht repariert werden, treiben Adaption an</h4>
 
 W&#xFC;rde es nicht besser sein, wenn die MMR nie eine Fehlpaarung &#xFC;bersehen w&#xFC;rde? Auch niedrige Mutationsraten k&#xF6;nnen ein Problem f&#xFC;r einen Organismus darstellen. Mutationen tragen n&#xE4;mlich auch zur genetischen Variation in einer Population bei. Zum Beispiel kann ein freiz&#xFC;giges Mismatch-Reparatursystem in einem Bakterium zuf&#xE4;llig zur Mutation eines Gens f&#xFC;hren, das eine Antibiotikaresistenz verleiht, wodurch die &#xDC;berlebens -und Reproduktionschancen der Bakterien in der Gegenwart von Antibiotika erh&#xF6;ht werden. Dies ist eine gute Nachricht f&#xFC;r die Bakterienpopulation, aber eine schlechte Nachricht f&#xFC;r Menschen, die zur Bek&#xE4;mpfung von Infektionskrankheiten auf Antibiotika angewiesen sind.
 
 Tats&#xE4;chlich erlangen Staphylococcus aureus Arten zunehmend eine Multidrug-Resistenz, was bedeutet, dass wenige oder keine Antibiotika die Ausbreitung dieses Bakteriums in einem Patienten verhindern k&#xF6;nnen. Infektionen mit multiresistenten Bakterien sind beim Menschen mit einer hohen Sterblichkeitsrate verbunden. Der weit verbreitete Einsatz von Antibiotika in der Tierproduktion und eine unangemessen verk&#xFC;rzte Verabreichung von Antibiotika tragen zur Entstehung multiresistenter Bakterien bei.

Mismatch repair helps fix DNA copying errors after replication. When DNA is copied, small mistakes can slip through. This repair system finds those mismatches and corrects them before they become permanent changes.

The process depends on a group of mutator proteins. These proteins work together to detect the error, remove the incorrect section, and help replace it with the proper DNA sequence. In this way, mismatch repair supports the accuracy of genetic information.

If the repair system does not work well, the mistake can remain in the DNA. That can raise the chance of mutation over time. For this reason, mismatch repair is an important safeguard during DNA replication.
