Membrane Fluidity

Die Membranfluidität ist die Fähigkeit von Phospholipiden und Proteinen, sich frei in der Membran zu bewegen. Es wird von den Phospholipidtypen in der Membran, der Temperatur der Umgebung und der Cholesterinkonzentration beeinflusst.
Gesättigte Phospholipide haben gerade Fettsäureketten ohne Doppelbindungen, während ungesättigte Phospholipide eine Biegung in ihren Fettsäureketten mit mindestens einer Doppelbindung aufweisen. 
Gesättigte Phospholipide bilden eine dicht gepackte Membran, was zu einer geringeren Membranfluidität führt. Ungesättigte Phospholipide stören die Phospholipidpackung, was zu einer höheren Membranfluidität führt.
Bei niedrigen Temperaturen sind die Membranlipide dicht gepackt und bewegen sich wenig, was zu einer starren Membran führt. Die Lipide zeigen bei hohen Temperaturen eine erhöhte Bewegung, wodurch die Membran flüssiger wird.
Cholesterin, eine weitere Membrankomponente, wird zwischen die Phospholipide eingefügt, wodurch ein Raum entsteht, der die Fließfähigkeit bei kälteren Temperaturen erhöht.
Bei wärmeren Temperaturen immobilisieren die Steroidringe im Cholesterin die Phospholipide und verhindern, dass die Membran zu flüssig wird.
Organismen halten unter verschiedenen Bedingungen eine optimale Membranfluidität aufrecht, indem sie das Verhältnis von gesättigten zu ungesättigten Phospholipiden und der Cholesterinkonzentration regulieren.

Die Membranfluidität wird durch das Fluid-Mosaik-Modell der Zellmembran beschreibt, das die Plasmamembranstruktur als ein Mosaik aus Komponenten – darunter Phospholipide, Cholesterin, Proteine und Kohlenhydrate – , die der Membran einen fluiden Charakter verleihen.

Mosaikcharakter der Membran

Die Mosaikcharakteristik der Membran trägt dazu bei, dass die Plasmamembran fluide bleibt. Die integralen Proteine und Lipide liegen als separate, aber lose verbundene Moleküle in der Membran vor. Die Membran ist eine relativ starre Struktur, die platzen kann, wenn sie durchstochen wird oder wenn die Zelle zu viel Wasser aufnimmt. Aufgrund ihrer mosaikartigen Beschaffenheit kann eine sehr feine Nadel jedoch leicht die Membran durchdringen, ohne dass diese platzt; die Membran fließt und schließt sich wieder, wenn die Nadel herausgezogen wird.

Natur der Phospholipide

Als nächstes geht es um die Natur der Phospholipide, aus denen die Lipiddoppelschicht besteht; In gesättigter Form sind die Fettsäuren in den Phospholipidschwänzen mit gebundenen Wasserstoffatomen gesättigt, was zu geraden Schwänzen führt. Im Gegensatz dazu enthalten ungesättigte Fettsäuren einige Doppelbindungen zwischen benachbarten Kohlenstoffatomen, was zu einer Biegung der Kohlenstoffkette um etwa 30 Grad führt. Wenn ungesättigte Fettsäuren komprimiert werden, drücken die „Knicke“ in ihren Schwänzen die benachbarten Phospholipidmoleküle weg und sorgen so dafür, dass zwischen den Phospholipidmolekülen etwas Platz bleibt. Dieser „Ellenbogenraum“ trägt dazu bei, die Fluidität der Membran bei Temperaturen aufrechtzuerhalten, bei denen Membranen mit gesättigten Fettsäureschwänzen in ihren Phospholipiden „einfrieren“ oder sich verfestigen würden. Die relative Fließfähigkeit der Membran ist in einer kalten Umgebung entscheidend. Eine kalte Umgebung neigt dazu, Membranen, die größtenteils aus gesättigten Fettsäuren bestehen, zu komprimieren, wodurch sie weniger flüssig und anfälliger für Risse werden.

Cholesterin

Bei Tieren ist Cholesterin ein weiterer Faktor, der zur Aufrechterhaltung der Membranfluidität beiträgt. Es liegt neben den Phospholipiden in der Membran vor und verringert tendenziell die Temperatureinwirkung auf die Membran. Somit fungiert Cholesterin als Puffer, der verhindert, dass niedrigere Temperaturen die Fluidität beeinträchtigen und höhere Temperaturen die Fluidität zu sehr erhöhen. Cholesterin erweitert in beide Richtungen den Temperaturbereich, in dem die Membran entsprechend fluide und funktionell aktiv ist. Cholesterin erfüllt auch andere Funktionen, beispielsweise die Organisation von Clustern von Transmembranproteinen in Lipid-Rafts. Durch die Fluidität der Membran können sich Proteine und Lipide bewegen und an bestimmten Stellen ansammeln, um Lipid-Rafts und interzelluläre Verbindungen zu bilden und so die Membranpermeabilität aufrechtzuerhalten.

how cell membranes stay flexible

Cell membranes stay flexible because of the fluid mosaic model. The plasma membrane is a mosaic of phospholipids, cholesterol, proteins, and carbohydrates. These parts are separate but loosely attached, so the membrane keeps a fluid character. The membrane is still fairly rigid, but it can flow and self-seal after a very fine needle passes through it.

Phospholipid tails also affect membrane fluidity. Saturated fatty acids in phospholipids have straight tails because they are packed with hydrogen atoms. Unsaturated fatty acids have double bonds that create a bend, or kink, of about 30 degrees in the carbon chain. When these bent tails are compressed, they push nearby molecules apart and leave enough space to help the membrane stay fluid.

This extra space matters in cold conditions. Membranes with mostly saturated fatty acid tails can become less fluid, and they may freeze, solidify, or rupture more easily in the cold. Unsaturated tails help prevent that by keeping the membrane from packing too tightly. In this way, membrane fluidity supports cell survival in cooler environments.

In animal cells, cholesterol also helps control membrane fluidity. It sits beside the phospholipids and reduces the effect of temperature changes. Cholesterol acts like a buffer, so low temperatures do not slow fluidity too much and high temperatures do not make the membrane too fluid. It widens the temperature range in which the membrane stays active and also helps organize clusters of transmembrane proteins into lipid rafts. Membrane fluidity also allows proteins and lipids to move and gather at places where lipid rafts and intercellular junctions form, which helps maintain membrane permeability.
