Membrane Lipids

In der Membran sind drei Arten von Lipiden vorhanden – Phosphoglyceride, Sphingolipide und Sterole.
Phosphoglyceride, auch bekannt als Glycerophospholipide, enthalten ein Glycerin-Rückgrat mit einer hydrophilen Phosphatgruppe und zwei hydrophoben Fettacylketten, die normalerweise 16 bis 22 Kohlenstoffatome lang sind.
Einige der häufigsten Phosphoglyceride in der Zellmembran sind Phosphatidylcholin, Phosphatidylethanolamin, Phosphatidylserin und Phosphatidylinositol - gebildet durch die Bindung von Cholin, Ethanolamin, Serin und Inositol an die Phosphatgruppe.
Sphingolipide sind Lipide, die ein Sphingosin-Rückgrat enthalten. Sphingosin ist ein Aminoalkohol mit einer langen, ungesättigten Kohlenwasserstoffkette. 
Das einfachste Sphingolipid ist Ceramid, das entsteht, wenn ein Fettsäuremolekül an die Aminogruppe des Sphingosins bindet.
Die verbleibende Hydroxylgruppe von Ceramid kann Ester mit anderen chemischen Gruppen wie Phosphocholin oder Kohlenhydraten bilden, wodurch Sphingomyelin bzw. Glykolipide gebildet werden.
Die dritte Art von Membranlipiden, Sterole, wie z. B. Cholesterin, kann keine eigenständige Lipiddoppelschicht bilden. Stattdessen interkalieren sie in Phospholipid-Doppelschichten und fungieren als Fließpuffer für die Membran.

Lipide sind ein wesentlicher Bestandteil aller biologischen Membranen. Der durchschnittliche Lipidgehalt in Membranen von Säugetieren beträgt 50 %, in der inneren Mitochondrienmembran kann er jedoch nur 20 % und in der die Nervenzellen umgebenden Myelinscheide bis zu 80 % betragen.

Phosphatidylcholin, Phosphatidylethanolamin, Phosphatidylserin und Sphingomyelin sind die am häufigsten bei Säugetieren vorkommenden Membranphospholipide. Bei physiologischem pH-Wert ist Phosphatidylserin negativ geladen, während die anderen drei neutral bleiben. In Membranlipiden können die Anzahl der in den Fettsäuren vorhandenen Kohlenstoffatome und ihr Sättigungsgrad variieren. Die Fettsäuren können vollständig gesättigt ohne Doppelbindungen, einfach ungesättigt mit nur einer Doppelbindung oder mehrfach ungesättigt mit mehreren Doppelbindungen sein.

Glykolipide umfassen eine physiologisch wichtige Klasse von Membranlipiden, die immer auf der äußeren oder extrazellulären Seite der Membran vorhanden sind. Bei Eukaryoten kommen sie sowohl in der Plasmamembran als auch in den intrazellulären Membranen vor. Galactocerebrosid, ein einfaches Glykolipid, kommt zusammen mit Sphingomyelin reichlich in Myelinscheiden vor. Ganglioside, die komplexesten Glykolipide, sind aufgrund der Anwesenheit von Sialinsäure negativ geladen und können bis zu 10 % der gesamten Lipide in Nervenzellmembranen ausmachen. Ganglioside werden auf ihre Rolle bei viralen und genetischen Erkrankungen untersucht. Das Gangliosid GM1 fungiert als Zelloberflächenrezeptor für das Choleratoxin, während bei der Tay-Sachs-Krankheit, einer genetischen Störung, eine übermäßige Anreicherung von GM2-Gangliosiden beobachtet wird.

Eine weitere Klasse von Membranlipiden sind die Sterole. Pflanzenspezifische Sterole stellen einen wesentlichen Bestandteil der pflanzlichen Plasmamembran dar und können 30–50 % der gesamten Membranlipide ausmachen. In eukaryotischen Membranen sind auch Cholesterine vorhanden, die fast 50 % der gesamten Lipide ausmachen. Die Struktur von Cholesterin in menschlichen Zellmembranen unterscheidet sich geringfügig von der von Ergosterin, einem häufigen Lipidbestandteil der Pilzmembranen.

Membrane lipids give biological membranes their structure and help set their properties. In mammalian cells, lipids make up about 50% of the membrane on average. The amount can be much lower in the inner mitochondrial membrane or much higher in the myelin sheath around nerve cells.

Several phospholipids are common in mammalian membranes. These include phosphatidylcholine, phosphatidylethanolamine, phosphatidylserine, and sphingomyelin. At physiological pH, phosphatidylserine carries a negative charge, while the other three are neutral.

The fatty acids in membrane lipids can differ in carbon length and in saturation. Some are fully saturated, which means they have no double bonds. Others are monounsaturated with one double bond, or polyunsaturated with multiple double bonds.

Glycolipids are an important membrane lipid class that stays on the outer, or extracellular, side of the membrane. In eukaryotes, they are found in both the plasma membrane and intracellular membranes. Galactocerebroside is a simple glycolipid found in large amounts in myelin sheaths along with sphingomyelin.

Gangliosides are the most complex glycolipids. They are negatively charged because they contain sialic acid, and they can make up as much as 10% of the total lipids in nerve cell membranes. Their roles are also studied in viral and genetic diseases. GM1 acts as a cell surface receptor for cholera toxin, and excess GM2 ganglioside is seen in Tay-Sachs disease.

Sterols are another major class of membrane lipids. Plant-specific sterols can make up 30% to 50% of the total lipids in the plant plasma membrane. In eukaryotic membranes, cholesterol is also common and can account for almost 50% of the total lipids. Cholesterol in human cell membranes is slightly different from ergosterol, which is a common lipid in fungal membranes.
