Lipids as Anchors

Die drei gebräuchlichsten Arten von Lipidankern sind Prenylgruppen, Fettacylgruppen und Glykosylphosphatidylinositol- oder GPI-Anker.
Die Prenylgruppen sind die 15 Kohlenstoff-Farnesyl- und die 20 Kohlenstoff-Geranylgeranyl-Gruppen. Sie binden an Cysteinreste am oder in der Nähe des Carboxy-Endes von Proteinen. 
Gesättigte Myristinsäure mit 14 Kohlenstoffatomen und Palmitinsäure mit 16 Kohlenstoffatomen sind die häufigsten Fettacylanker, die mit Proteinen verbunden sind.
Die Zugabe von Myristinsäure oder Myristoylierung erfolgt am N-terminalen Glycinrest eines Proteins. Die Palmitoylierung der Proteine kann am N- oder C-terminalen Cysteinrest erfolgen.
Prenylierte, myristoylierte und palmitoylierte Proteine spielen eine wesentliche Rolle bei intrazellulären Signalwegen und Protein-Protein-Interaktionen.
GPI-Anker binden an extrazelluläre Proteine und enthalten eine Kernstruktur aus Phosphatidylinositol, Glucosamin, drei Mannosen und Phosphoethanolamin.
Das Phosphoethanolamin bindet sich an die C-terminale Aminosäure des Zielproteins, und das Phospholipid fügt sich in die äußere Schicht der Membran ein.
Dies ermöglicht es GPI-verankerten Proteinen, an extrazellulären Funktionen wie Zell-Zell-Kommunikation und Zelladhäsion teilzunehmen.

Prenyl- und Fettsäuregruppen fungieren als Anker auf der zytosolischen Oberfläche der Membran, während GPI Proteine auf der extrazellulären Seite verankert.

Der Carboxylterminus der meisten prenylierten Proteine, wie z. B. der Ras-Proteine, enthält das CaaX-Motiv oder die CaaX-Box, wobei „C“ für Cystein, „a“ für eine beliebige aliphatische Aminosäure und „X“ für jeden andere Aminosäurerest steht. Die an der Position „X“ vorhandene Aminosäure bestimmt, ob es sich bei der Bindung an das Protein um die Farnesyl- oder die Geranylgeranyl-Bindung handelt. Wenn das „X“ Alanin, Serin, Methionin, Cystein oder Glutamin ist, erkennt die Farnesyltransferase das CaaX-Motiv und verknüpft Farnesyl mit dem Protein. Wenn andererseits das „X“ Glutaminsäure oder Leucin ist, dann identifiziert die Geranyltransferase Typ I das CaaX-Motiv und bindet Geranylgeranyl daran. Nach der Prenylierung wird das Tripeptid aaX durch spezifische Proteasen entfernt und eine Methyltransferase methyliert den resultierenden negativ geladenen Cysteinrest. Durch Methylierung wird die negative Ladung des Cysteins entfernt, wodurch es hydrophob wird.

Manchmal wird mehr als ein Lipidanker verwendet, um Proteine an die Membran zu binden. Beispielsweise wird die zytoplasmatische Tyrosinkinase mit Hilfe von Myristin- und Palmitinsäure in der Membran verankert. Allerdings benötigen GPI-verankerte Proteine normalerweise keinen weiteren Anker zur Stabilisierung. Ein Ende des GPI enthält die beiden Fettsäuregruppen, die in die Lipiddoppelschicht eingefügt werden können. Lipid anchors help attach proteins to the plasma membrane. In eukaryotic cells, the main lipid anchors are prenyl groups, fatty acyl groups, and glycosylphosphatidylinositol, or GPI, groups. Prenyl and fatty acyl groups anchor proteins on the cytosolic side of the membrane. GPI anchors proteins on the extracellular side.

Prenylated proteins often have a CaaX motif at the carboxy terminus. In this sequence, C means cysteine, a means any aliphatic amino acid, and X means any other amino acid residue. The amino acid at the X position helps decide the type of linkage. If X is alanine, serine, methionine, cysteine, or glutamine, farnesyl transferase attaches a farnesyl group. If X is glutamic acid or leucine, geranylgeranyl transferase type I attaches a geranylgeranyl group.

After prenylation, specific proteases remove the aaX tripeptide. Then a methyltransferase methylates the cysteine that remains. This step removes the negative charge on the cysteine and makes it more hydrophobic. The change helps the protein stay associated with the membrane.

Some proteins use more than one lipid anchor. A cytoplasmic tyrosine kinase, for example, is anchored with both myristic acid and palmitic acid. GPI-anchored proteins usually do not need a second anchor for stability. The GPI group has two fatty acyl groups that insert into the lipid bilayer. Its other end contains phosphoethanolamine, which links to target proteins involved in different cell functions.
