Protein Diffusion in the Membrane

Bei physiologischen Temperaturen wirkt eine Lipiddoppelschicht wie eine zweidimensionale Flüssigkeit, die die Diffusion von Proteinen, Lipiden und anderen Membrankomponenten ermöglicht.
Bei der lateralen Proteindiffusion handelt es sich um eine Seitwärtsbewegung innerhalb der Membran.
Die extrazelluläre Matrix kann mit der extrazellulären Domäne eines Proteins interagieren und seine Bewegung stören, oder intrazellulär können das Aktin-Zytoskelett und die zugehörigen Transmembranproteine Zäune bilden, was zur Bildung spezifischer Kompartimente führt, die als Korallen bekannt sind.

Die FRAP-Technik (fluorescence recovery after photobleaching) kann verwendet werden, um die Diffusionsrate von Membranproteinen zu bestimmen. Bei dieser Methode wird das Zielprotein durch einen mit einem Fluorophor verbundenen Antikörper markiert, der für das Protein spezifisch ist. Abhängig von der Position und Struktur des Proteins hat jedes Protein einen anderen Diffusionskoeffizienten.

  Die FRAP-Studien zeigten, dass nicht alle Membranproteine gleich mobil sind und die Diffusionsrate von Membranproteinen langsamer ist als in einer reinen Lipiddoppelschicht erwartet werden kann. Die Diffusionsgeschwindigkeit von Proteinen durch die Membran ist geringer als die Diffusionsgeschwindigkeit von Membranlipiden. Um beispielsweise die 20 Mikrometer betragende Länge einer eukaryotischen Zelle zu durchqueren, benötigen Membranlipide etwa 20 Sekunden, während Membranproteine bis zu 600–3600 Sekunden benötigen können. Neben der FRAP wurden auch andere fortgeschrittene Techniken wie 1) Einzelpartikelverfolgung, 2) der Einsatz von optischen Pinzetten zur Untersuchung von Membranbarrieren und 3) genetisch veränderte Proteine zur Untersuchung der Rolle der zytoplasmatischen und exoplasmatischen Proteinbereiche bei der lateralen Diffusion eingesetzt, um verschiedene Aspekte der Proteindiffusion in der Membran zu erforschen.

Membrane proteins can move across the cell surface by lateral diffusion, and they can also rotate in place. Scientists confirmed lateral movement with a cell fusion experiment. Mouse and human cells were fused to make hybrid cells. The membrane proteins on the two cells were tagged with red and green fluorescent markers. At first, each color stayed on its own side of the fused cell. After 40 minutes, the colors mixed across the whole cell surface, showing that the proteins had moved laterally.

FRAP, or fluorescence recovery after photobleaching, is another way to measure membrane protein diffusion. In this method, the target protein is labeled with a fluorophore-linked antibody that binds only to that protein. A genetically engineered protein fused to green fluorescent protein can also be used. A laser then creates a bleached, non-fluorescent area. If 40% of the fluorescence returns, about 40% of the proteins in that area are considered laterally diffusible.

The time needed for recovery helps scientists calculate the diffusion rate and the diffusion coefficient. Different proteins have different diffusion coefficients because their location and structure are not the same. FRAP studies also showed that membrane proteins move more slowly than expected from the pure lipid bilayer. Their diffusion is slower than membrane lipids. For example, lipids may cross the 20-micrometer length of a eukaryotic cell in about 20 seconds, while membrane proteins may take 600 to 3600 seconds.

Researchers also use other advanced methods to study membrane protein movement. These include single-particle tracking, optical tweezers to study membrane barriers, and genetically modified proteins that help show how cytoplasmic and exoplasmic domains affect lateral diffusion.
