The Y Chromosome Determines Maleness

Beim Menschen gibt es zwei Geschlechtschromosomen: X und Y.
Das Y-Chromosom bestimmt das männliche Geschlecht und ist das kleinste Chromosom des menschlichen Genoms.
Es gibt drei Regionen im Y-Chromosom: den pseudoautosomalen Teil, der weiter in zwei Regionen unterteilt ist - PAR1 und PAR2, die heterochromatische Region und die euchromatische Region.
Das S-R-Y-Gen befindet sich im euchromatischen Bereich des Y-Chromosoms. Es spielt eine wichtige Rolle bei der Entwicklung männlicher Föten.
Wenn eine Eizelle, die ein X-Chromosom trägt, mit einem Spermium mit einem Y-Chromosom verschmilzt, bildet sie eine Zygote.
Die Zygote teilt sich dann kontinuierlich und bildet einen Embryo. In diesem Stadium entwickeln sich männliche und weibliche Embryonen auf dem gleichen Weg und sind physiologisch identisch.
In den frühen Entwicklungsstadien hat der Embryo zwei Vorläuferorgane: den Wolffschen Gang und den Müller-Gang.
Der Wolffsche Gang kann sich in männliche Genitalstrukturen differenzieren und der Müllersche Gang in weibliche Genitalstrukturen.
In den Wochen nach der Befruchtung wird das auf dem Y-Chromosom vorhandene S-R-Y-Gen transkribiert und dann in das S-R-Y-Protein übersetzt. Das S-R-Y-Protein initiiert die Entwicklung der Hoden beim Fötus.
 Das S-R-Y-Protein fungiert als Transkriptionsfaktor und bindet an bestimmte Stellen auf der DNA, wodurch der Hodenbildungsweg aktiviert wird.
Dann beginnen die Keimdrüsen, mehrere Hormone auszuschütten, darunter das Anti-Müller-Hormon und Testosteron.
Das Anti-Müller-Hormon hemmt die Entwicklung des Müller-Gangs.
Testosteron hingegen initiiert die Entwicklung des Wolffschen Ganges in Samenleiter, Ejakulationsgänge und Samenbläschen.
Wenn dieser Signalweg gestört ist und die Gonaden nicht in der Lage sind, Testosteron und das Anti-Müller-Hormon zu sezernieren, entwickelt sich der Müller-Gang zu weiblichen Fortpflanzungsorganen, selbst wenn der Fötus genotypisch XY ist.

Das Y-Chromosom ist ein Geschlechtschromosom, das bei mehreren Wirbeltieren und Säugetieren, einschließlich des Menschen, vorkommt. Neben 22 Paaren von Autosomen haben menschliche Männer ein X-Chromosom und ein Y-Chromosom. Bei diesen Organismen bestimmt das Vorhandensein oder Fehlen des Y-Chromosoms die Entwicklung männlicher Merkmale.

Evolution

Vor etwa 300 Millionen Jahren spalteten sich die beiden Geschlechtschromosomen von zwei identischen autosomalen Chromosomen ab. Im Laufe der Zeit hat das Y-Chromosom die meisten seiner Gene verloren und ist in der Größe geschrumpft. Heute teilt nur ein kleiner Teil des Y-Chromosoms Sequenzähnlichkeiten mit dem X-Chromosom. Eine umfangreiche wissenschaftliche Studie über Y-Chromosomen bei acht Säugetieren, darunter Affen, Mäuse, Ratten, Stiere, Opossums, Schimpansen und Menschen, zeigte 18 hochgradig ähnliche Regionen auf Y-Chromosomen. Die Tatsache, dass diese Regionen bei all diesen Säugetieren erhalten geblieben sind, trotz des Verlusts eines großen Teils der Y-Chromosomen während der Evolution, zeigt, dass die Gene auf den Y-Chromosomen lebenswichtig für das Überleben dieser Säugetiere sind.

Struktur

Das Y-Chromosom der Säugetiere hat drei Hauptregionen. Die pseudoautosomalen Regionen auf dem Y-Chromosom teilen Homologie mit dem X-Chromosom und können meiotische Rekombinationen durchlaufen. Daher werden Gene, die in den pseudoautosomalen Regionen vorhanden sind, in autosomaler und nicht in geschlechtsgebundener Weise vererbt. Die zweite Region ist eine Euchromatinregion, die als männlich-spezifische Y-Chromosom-Region oder MSY bezeichnet wird. Sie erstreckt sich über 23 Mb Länge und enthält einige protein-kodierende Gene, wie das SRY-Gen, einen Hauptregulator der Männlichkeit, und das ZFY-Gen, das für die Spermatogenese benötigt wird. Die dritte Region ist eine Heterochromatinregion, die mehrere repetitive Sequenzen enthält.

Mutationen und ihre Auswirkungen

Deletionen in der MSY-Region des Y-Chromosoms beeinträchtigen die Spermienentwicklung und verursachen männliche Sterilität. The Y chromosome is a sex chromosome that helps guide male development in humans and other vertebrates and mammals. Human males usually have one X chromosome and one Y chromosome, along with 22 pairs of autosomes. In these organisms, the presence or absence of the Y chromosome influences whether male traits develop.

The Y chromosome evolved from an autosome about 300 million years ago, when two sex chromosomes began to diverge. Since then, it has lost most of its genes and become much smaller. Only a small region still shares sequence similarity with the X chromosome. A large study of Y chromosomes in eight mammals, including monkeys, mice, rats, bulls, opossums, chimpanzees, and humans, found 18 highly similar regions. Their long-term retention suggests that Y-linked genes remain important for mammalian survival.

The mammalian Y chromosome has three main regions. The pseudoautosomal regions match parts of the X chromosome and can undergo meiotic recombination, which means they can be inherited more like autosomes than sex-linked genes. The second region is the male-specific Y chromosome region, or MSY. It is a euchromatin region about 23 Mb long and contains few protein-coding genes, including SRY, a master regulator of maleness, and ZFY, which is required for spermatogenesis. The third region is heterochromatin, which contains many repetitive sequences.

Changes in the MSY region can have major effects on reproduction. Deletions in this region can disrupt sperm development and lead to male sterility. For example, deletions in the AZF gene on the long arm of the Y chromosome can cause azoospermia, a condition in which spermatozoa are not found in the male ejaculate. Deletions in specific MSY regions are also linked to testicular germ cell tumors, or TGCTs.
