Single-Strand DNA Binding Proteins

Sobald die Helikase die DNA abwickelt, können die daraus resultierenden getrennten, einzelsträngigen DNA-Moleküle Haarnadelschleifen durch Intrastrangpaarung oder doppelsträngige DNA durch Interstrangpaarung bilden. 
Diese Stränge sind auch anfällig für Angriffe von Nukleasen, die die DNA verdauen können.
Um dies zu verhindern, binden Einzelstrang-DNA-bindende Proteine (SSB) sequentiell an die einzelsträngige DNA, wobei das eingehende SSB neben dem vorhandenen SSB bindet.
Diese kooperative Bindung der SSB-Proteine begradigt und schützt den DNA-Elternstrang und verhindert die Bildung der Haarnadelschleifen oder das Zurückspulen der DNA in ihre doppelsträngige Struktur.
Die SSB-Proteine binden fest an das Zucker-Phosphat-Rückgrat der DNA und lassen die stickstoffhaltigen Basen für die komplementäre Nukleotidbindung verfügbar, was die Bildung des Tochterstrangs ermöglicht.

Für eine erfolgreiche DNA-Replikation muss das Entwinden der doppelsträngigen DNA von der Stabilisierung und dem Schutz der getrennten Einzelstränge der DNA begleitet sein. Diese entscheidende Aufgabe wird von Einzelstrang-DNA-bindenden (SSB) Proteinen übernommen. Sie binden an die DNA in einer sequenzunabhängigen Weise, was bedeutet, dass die Stickstoffbasen der DNA nicht in einer spezifischen Reihenfolge vorhanden sein müssen, damit die SSB-Proteine an sie binden können. Die Bindung von SSB-Proteinen richtet einzelsträngige DNA (ssDNA) aus und macht sie steif. Es wird angenommen, dass dies die Fähigkeit der DNA-Polymerase verbessert, Basen korrekt auszuwählen, wodurch die Genauigkeit der DNA-Replikation erhöht wird.

Single-strand DNA-binding proteins, or SSB proteins, help protect DNA during replication. When double-stranded DNA is unwound, the two strands separate. SSB proteins bind to the exposed single strands and keep them stable.

These proteins bind in a sequence-independent manner. That means the order of the nitrogenous bases does not have to follow a specific pattern for SSB proteins to attach. Their binding straightens single-stranded DNA and makes it more rigid.

This support may help DNA polymerase choose the correct bases during copying. As a result, DNA replication can become more accurate, or more faithful. Because SSB proteins also take part in DNA recombination and repair, researchers are studying them as possible antibiotic targets against drug-resistant microorganisms.
