Biomoleküle wie Proteine und Nukleinsäuren sind schwerflüchtige Verbindungen mit hohen Molekulargewichten. Die konventionelle Elektronenstoßionisations-Massenspektrometrie steht bei solchen Molekülen vor Herausforderungen, einschließlich einer starken molekularen Fragmentierung.
Die Elektrospray-Ionisation ist eine alternative "weiche" Ionisationsmethode, die nichtradikale Molekülionen für die Massenspektrometrie solcher Biomoleküle erzeugt.
Bei diesem Verfahren wird die Probe mit einem polaren Lösungsmittel wie Methanol, Methanol-Wasser oder Acetonitril mit oder ohne Zusatz von Salzen gemischt.
Das Gemisch wird über eine Hochspannungskapillare in eine Vakuumkammer gesprüht. Die schnelle Verdunstung des Lösungsmittels aus dem feinen Sprühnebel geladener Tröpfchen führt schließlich zum geladenen gasförmigen Probenmolekül.
Dieses geladene Probenmolekül wird im Massenspektrum als Summe der Molekülmasse der Probe und der Massen der zugehörigen Ionen detektiert.
Wenn beispielsweise Natriumionen in der ionischen Lösung vorhanden sind, weist das Spektrum typischerweise einen M+23-Peak auf.

Biomoleküle mit höherem Molekulargewicht sind nichtflüchtige Verbindungen, die sich bei der Massenanalyse mit herkömmlicher Elektronenstoßionisation vor der Ionisierung oder Verdampfung zersetzen können. Dementsprechend ist die Elektrospray-Ionisation (ESI) die bevorzugte Methode zum Verdampfen und Ionisieren von Biomolekülen, da sie eine schnelle Fragmentierung umgeht und die Aufzeichnung von Massensignalen für das gesamte Biomolekül ermöglicht.

ESI nutzt elektrische Energie, um Ionen aus der flüssigen Phase der Probe in die gasförmige Phase zu übertragen. Das Analyt-Biomolekül wird mit einer ionischen Flüssigkeit vorgemischt – einer nichtmolekularen Verbindung, die als Ionisierungsquelle dient. Die Lösung wird durch eine Hochspannungskapillare gesprüht, um ein feines Aerosol zu erzeugen. Der zwischen der Kapillare und dem Eingang der Probekammer angelegte Spannungsunterschied bewirkt die Bewegung geladener Tröpfchen in Richtung der Probekammer. Die ionische Flüssigkeit aus dem Aerosol verdampft während dieser Bewegung und hinterlässt ein unterschiedlich protoniertes oder ionisiertes Analytmolekül. Diese geladenen Spezies werden im Massenspektrum bei einem m/z-Wert nachgewiesen, der der Summe der Molekularmasse des Analytmoleküls und des Molekulargewichts der Ionen aus der mit dem Analytmolekül verbundenen ionischen Flüssigkeit entspricht.

Die Position der Massensignale variiert je nach Art der in der ionischen Flüssigkeit vorhandenen Kationen. Beispielsweise führt die Anwesenheit von Natriumionen in der ionischen Flüssigkeit zu einem (M+23)-Peak in den Massenspektren, während die Anwesenheit eines Protons zu einem (M+1)-Peak führt.

Electrospray ionization, or ESI, is a mass spectrometry method used to detect biomolecules that are too large or nonvolatile for conventional electron impact ionization. These molecules may break down before they can ionize or evaporate. ESI helps prevent rapid fragmentation and lets scientists record a mass signal for the whole biomolecule.

ESI uses electrical energy to move ions from the liquid phase into the gas phase. The analyte biomolecule is mixed with an ionic liquid, which is a nonmolecular compound that acts as the ionization source. The solution then passes through a high-voltage capillary and forms a fine aerosol.

A voltage difference between the capillary and the entrance to the analysis chamber pulls the charged droplets toward the chamber. As the droplets travel, the ionic liquid evaporates. This process leaves behind a protonated or otherwise ionized analyte molecule.

The charged species are measured in the mass spectrum by their m/z value, which reflects the analyte’s molecular mass plus the mass of the ions from the ionic liquid attached to it. The peak position depends on which cations are present in the ionic liquid. For example, sodium ions produce an (M+23) peak, while a proton produces an (M+1) peak.
