Die α Wasserstoffatome der Carbonyle sind von Natur aus sauer, da ihre konjugierten Basen resonanzstabilisiert sind. Daher sind Enolationen aufgrund ihrer Lösungsstabilität und besseren Nukleophilität bei Reaktionen oft nützlicher als Enole.
Wie das allylische Anion verhält sich das Enolat-Ion wie ein konjugiertes System mit drei Atomen und vier Elektronen. Der Ersatz von Kohlenstoff durch Sauerstoff senkt die Energie der beiden besetzten π Molekülorbitale und verzerrt sie.
Im am tiefsten besetzten π Orbital werden die Elektronen über die drei Atome delokalisiert, wobei der elektronegative Sauerstoff einen größeren Beitrag leistet. Diese zusätzliche Überlappung und Stabilisierung macht Carbonyl- α Wasserstoffatome saurer als allylische Wasserstoffatome von Alkenen.
Der HOMO hat einen signifikanten α-Kohlenstoff-Charakter mit einem Knoten an der Kohlenstoff-Sauerstoff-Bindung. Während Reaktionen, die durch Wechselwirkung mit dem HOMO dominiert werden, tendenziell am α Kohlenstoff ablaufen und zu einer α-Substitution führen, neigen Reaktionen, die durch elektrostatische Wechselwirkungen angetrieben werden, dazu, auf Sauerstoff abzulaufen und Enolderivate zu bilden.
Da Enolate zwei unterschiedliche reaktive nukleophile Zentren haben, handelt es sich um ambidente Nukleophile.

Enolationen entstehen durch die Säure-Base-Reaktion einer Carbonylverbindung mit einer Base. Dies führt zur Deprotonierung des α-Wassersubstanzatoms, was zu einem resonanzstabilisierten Enolation führt, bei dem eine der beitragenden Strukturen ein Oxyanion ist, das zusätzliche Stabilität verleiht. Daher ist das Proton am α-Kohlensubstanz von Natur aus saurer als das anderer sp^3-hybridisierter CH-Bindungen, aber weniger saurer als das Proton in O-H-Bindungen, bei denen die negative Ladung in der konjugierten Base am Sauersubstanzatom lokalisiert ist. Dies spiegelt sich in ihrem Trend der pK_a-Werte wider. Beispielsweise haben Essigsäure, Ethanol, Aceton, 1-Propen und Ethan pK_a-Werte von 4,8, 16, 19,2, 43 bzw. 50.

Das Enolation ist ein Beispiel für ein ambidentes Nukleophil – also ein Nukleophil mit zwei reaktiven Zentren. Die beitragenden Strukturen der Enolationen zeigen, dass sowohl Kohlensubstanz- als auch Sauersubstanzatome die negative Ladung tragen können. Daher ist das Enolation die konjugierte Base sowohl der Keto- als auch der Enolform. Theoretisch kann es mit einem bestimmten Elektrophil reagieren und durch Bindungsbildung an den beiden verschiedenen Stellen zwei verschiedene Produkte bilden. Ein Enolation reagiert jedoch normalerweise am Kohlensubstanzende, da dieses nukleophiler ist als die Sauersubstanzstelle.

Da Enolationen Brønsted-Basen sind, reagieren sie wie Protonen mit Brønsted-Säuren. Dies führt zu einem Wassersubstanzaustausch an der α-Position von Carbonylverbindungen mit der des Lösungsmittels, was in Gegenwart von D_2O und einer wässrigen Base zu einem Isotopenaustausch führt. Ein optisch aktiver Aldehyd oder Keton unterliegt einer Racemisierung, wenn das Molekül einen asymmetrischen α-Kohlensubstanz enthält. Der Verlust an Stereogenität ist auf die Bildung eines achiralen Enolat-Zwischenprodukts zurückzuführen, bei dem alle drei Atome aufgrund der sp^2-Hybridisierung und Konjugation durch p-Orbitalüberlappung trigonal-planar sind. Da der pK_a eines α-Wassersubstanzen bei Estern sehr hoch ist, werden die verschiedenen Folgen der Enolat-Ionenbildung speziell bei Aldehyden und Ketonen beobachtet.

Enolationen reagieren auch als Lewis-Basen und wirken dort als Nukleophile. Daher können sie zwei Arten von Reaktionen eingehen, die zur Bildung neuer Bindungen am α-Kohlensubstanz führen:

Enolate ions form when a base removes the alpha hydrogen from a carbonyl compound. This acid-base reaction gives a resonance-stabilized ion, and one contributing form places the negative charge on oxygen. That oxygen-containing form adds extra stability.

The alpha hydrogen in a carbonyl compound is more acidic than other sp3 C-H bonds, but it is less acidic than O-H bonds. This difference appears in pKa values. Acetic acid has a pKa of 4.8, ethanol 16, acetone 19.2, 1-propene 43, and ethane 50.

Enolate ions are ambident nucleophiles, which means they have two reactive sites. Their resonance structures show that the negative charge can be on carbon or oxygen. So an enolate can, in theory, form different products by reacting at either site.

In practice, enolates usually react at the carbon end because it is more nucleophilic than the oxygen site. They also behave as Brønsted bases and can react with protons. This can cause hydrogen exchange at the alpha position with the solvent, including isotope exchange in D2O with aqueous base.

Optically active aldehydes or ketones can also racemize if they have an asymmetric alpha carbon. Racemization means loss of optical activity. It happens because the enolate intermediate is achiral, and the atoms around it are trigonal planar with sp2 hybridization and p-orbital overlap.

These effects are seen mainly with aldehydes and ketones, since alpha hydrogens in esters have very high pKa values. Enolates also act as Lewis bases and nucleophiles. They can form new bonds at the alpha carbon through substitution with electrophiles, such as halogenation with X2 in acid or base, alkylation with alkyl halides or sulfonate esters, and through addition to carbonyl groups followed by nucleophilic acyl substitution when the carbonyl structure allows it.
