Unsymmetrische Ketone mit nicht-äquivalenten α-Wasserstoffatomen ergeben zwei mögliche Zwischenprodukte – weniger substituierte oder mehr substituierte Enolate.
Da die Deprotonierung für ein weniger behindertes Proton einfacher ist, werden weniger substituierte Enolate vergleichsweise schneller gebildet. Diese werden als kinetische Enolate bezeichnet. Da die stärker substituierten Enolate eine stark substituierte Doppelbindung aufweisen, sind sie stabiler und werden oft als thermodynamische Enolate bezeichnet. Daher haben thermodynamische Enolate ein niedrigeres Energieniveau als kinetische Enolate.
Da die Energie, die zur Bildung thermodynamischer Enolate benötigt wird, höher ist, erfordert ihre Bildung eine längere Reaktionszeit.
Eine regioselektive Bildung der Enolate ist in Abhängigkeit von den Reaktionsbedingungen möglich.
Zur Erzeugung kinetischer Enolate werden sperrige nicht-nukleophile starke Basen in aprotischen Lösungsmitteln verwendet, um gezielt ungehinderte Protonen zu abstrahieren. Darüber hinaus verbietet die Aufrechterhaltung einer niedrigen Reaktionstemperatur die Äquilibrierung der beiden Enolate.
Im Gegensatz dazu begünstigen nichtsterisch gehinderte Basen in protischen Lösungsmitteln bei Raumtemperatur die Bildung des thermodynamischen Enolats.

Normalerweise werden die thermodynamischen Enolate aus dem stärker substituierten α-Kohlenstoffatom gebildet, während die kinetischen Enolate schneller durch Deprotonierung aus der weniger substituierten Position gebildet werden. Die thermodynamischen Enolate haben eine niedrigere Energie und sind daher stabiler. Allerdings ist die zur Bildung kinetischer Enolate erforderliche Energie geringer.

Diese Regioselektivität bei der Enolatbildung ist im Wesentlichen eine Säure-Base-Reaktion und wird durch verschiedene Faktoren wie Lösungsmittel, Base, Kation und Temperatur gesteuert. Protische Lösungsmittel und schwächere Basen begünstigen die Bildung thermodynamischer Enolate, während aprotische Lösungsmittel und stärkere Basen die kinetischen Enolate begünstigen. Thermodynamische Enolate werden bei höheren Temperaturen gebildet und haben aufgrund der höheren Energiebarriere längere Reaktionszeiten. Andererseits werden kinetische Enolate bei niedrigeren Temperaturen und kurzen Reaktionszeiten gebildet. Die Bedingungen, die thermodynamische Enolate begünstigen, begünstigen eine reversible Reaktion, was bei den Bedingungen für kinetische Enolat-Zwischenprodukte nicht der Fall ist.

Unsymmetrical ketones can form two enolates, and the reaction conditions decide which one appears. One enolate comes from the less substituted alpha-carbon, and the other comes from the more substituted alpha-carbon. These pathways are linked to acid-base chemistry and to regioselectivity, which means choosing one position in the molecule over another.

The enolate from the more substituted alpha-carbon is the thermodynamic enolate. It has lower energy, so it is more stable. The enolate from the less substituted position is the kinetic enolate. It forms faster because less energy is needed to remove that proton.

Solvent, base, cation, and temperature all affect which enolate forms. Protic solvents and weaker bases tend to favor the thermodynamic enolate. Aprotic solvents and stronger bases tend to favor the kinetic enolate. Higher temperatures and longer reaction times also favor thermodynamic control because the reaction can reverse and reach the lower-energy product. Lower temperatures with short reaction times favor the kinetic enolate because the intermediate does not have time to equilibrate.
