Cytoplasm

In eukaryotischen Zellen ist das Zytoplasma der Bereich zwischen der Plasmamembran und der Kernhülle.
Da prokaryotische Zellen keinen Zellkern haben, bezieht sich das Zytoplasma auf alles, was in der Plasmamembran eingeschlossen ist.
In beiden Zelltypen wird der zytoplasmatische Raum durch eine gelartige Matrix, das Zytosol, ausgefüllt.
Das Zytosol ist eine wässrige Lösung, die viele lösliche Ionen, kleine Moleküle und Makromoleküle enthält.
Das Zytoplasma eukaryotischer Zellen umfasst auch zahlreiche membrangebundene Organellen, die spezialisierte zelluläre Funktionen erfüllen, wie die Mitochondrien, das endoplasmatische Retikulum und der Golgi-Apparat.
Ein Gerüst aus Proteinfasern, das im Zytoplasma vorhanden ist – das Zytoskelett – gibt der Zelle ihre Form und Struktur. Es hilft auch beim intrazellulären Transport und bei der Zellmotilität.
Darüber hinaus finden auch viele zelluläre Funktionen außerhalb der Organelle, wie z. B. die Proteinsynthese, im Zytoplasma statt. Die wässrige Natur des Zytoplasmas erleichtert die Proteinfaltung, indem hydrophobe Aminosäure-Seitengruppen in den Proteinkern abgestoßen werden.
Einige Zellen speichern in ihrem Zytoplasma auch unlösliche Stoffwechselprodukte wie Glykogen in Form von Einschlüssen.

Das Zytoplasma besteht aus Organellen und einem Gerüst aus Proteingerüsten, dem Zytoskelett, das in einer wässrigen Lösung, dem Zytosol, suspendiert ist. Das Zytosol ist eine reichhaltige Brühe aus Wasser, Ionen, Salzen und verschiedenen organischen Molekülen.

Proteinfaltung und -fehlfaltung

Das Zytoplasma ist der Ort für mehrere zelluläre Prozesse, einschließlich der Proteinsynthese und -faltung. Die wässrige Beschaffenheit des Zytosols fördert die Proteinfaltung, sodass die hydrophoben Aminosäureseitenketten im Proteinkern vergraben sind, während die hydrophilen Aminosäuren dem Zytosol zugewandt sind. Allerdings können zelluläre Belastungen wie Alterung und Veränderungen des pH-Werts, der Temperatur oder der Osmolarität zu einer Fehlfaltung des Proteins führen. Fehlgefaltete Proteine können sich im Zytoplasma ansammeln und unlösliche Proteinaggregate bilden. Solche Aggregate sind an neurodegenerativen Erkrankungen wie Alzheimer und Parkinson beteiligt.

Zusammensetzung und Funktion des Zytoskeletts

Das eukaryotische Zytoskelett besteht aus drei Arten von filamentösen Proteinen – Mikrotubuli, Mikrofilamenten und Zwischenfilamenten.

Mikrotubuli, die größte Art von Filamenten, bestehen aus dem Protein Tubulin. Es handelt sich um dynamische Strukturen, die durch Hinzufügen oder Entfernen von Tubulinmolekülen an den Enden ihrer Stränge wachsen oder schrumpfen können. Sie verleihen der Zelle strukturelle Stabilität und fungieren als Transportwege für Proteine, Vesikel und bestimmte Organellen innerhalb der Zelle. Darüber hinaus spielen Mikrotubuli eine entscheidende Rolle bei der Zellteilung, indem sie ein Gerüst bereitstellen, das die Chromosomen zu den gegenüberliegenden Enden der Zelle führt.

Mikrofilamente oder Aktinfilamente sind kleinere Zytoskelett-Filamente, die aus einem Protein namens Aktin bestehen. Aktinproteine können sich schnell zusammensetzen und wieder zerlegen, um Filamente zu bilden. Daher ermöglichen sie die Beweglichkeit einzelliger Organismen wie Amöben oder die Wanderung weißer Blutkörperchen zu Infektionsherden. In Skelettmuskelzellen gleiten Aktinfilamente entlang Myosinfilamenten, um die Muskelkontraktion zu vermitteln.

Zwischenfilamente sind nicht so dynamisch wie Mikrotubuli oder Aktinfilamente. Wie die beiden anderen Filamentarten sorgen sie jedoch auch für die strukturelle Unterstützung der Zelle. Zwischenfilamente bestehen je nach Zelltyp aus verschiedenen Arten von Proteinen. Beispielsweise enthalten Zwischenfilamente in Haaren und Nägeln Keratin, während diese Filamente in Muskelzellen aus Desminen bestehen.

The cytoplasm is the cell’s internal space that holds organelles and the cytoskeleton. It is suspended in the cytosol, an aqueous fluid made of water, ions, salts, and many organic molecules. This fluid helps keep the cell organized and supports many cellular activities.

The cytoplasm is also where protein synthesis and protein folding take place. The watery cytosol helps proteins fold correctly. In a folded protein, hydrophobic amino acid side chains tend to stay buried in the core, while hydrophilic amino acids face the cytosol.

Cell stress can disrupt this process. Changes in pH, temperature, osmolarity, or aging can cause proteins to misfold. These misfolded proteins may collect in the cytoplasm and form insoluble aggregates. Such aggregates are linked to neurodegenerative disorders like Alzheimer’s and Parkinson’s disease.

The cytoskeleton is the protein framework inside the cytoplasm. It includes three filament types: microtubules, microfilaments, and intermediate filaments. Microtubules are the largest and are built from tubulin. They can grow or shrink by adding or removing tubulin at their ends. They help give the cell structural stability and serve as tracks for moving proteins, vesicles, and some organelles. They also help guide chromosomes to opposite ends of the cell during cell division.

Microfilaments, also called actin filaments, are smaller and are made of actin. They can assemble and disassemble quickly. This rapid change helps amoeba move and helps white blood cells migrate to infection sites. In skeletal muscle cells, actin filaments slide along myosin filaments to drive muscle contraction.

Intermediate filaments are less dynamic than microtubules or actin filaments, but they still provide support. They are made of different proteins depending on the cell type. In hair and nails, intermediate filaments contain keratin. In muscle cells, they are made of desmins.
