Multi-pass Transmembrane Proteins and &#946;-barrels

In Multipass-Membranproteinen bestimmt die Wechselwirkung zwischen mehreren Transmembrandomänen deren Struktur und Funktion.
G-Protein-gekoppelte Rezeptoren sind die größte Familie von Membranproteinen. Sie enthalten sieben Transmembran-Alpha-Helices, die Signale zwischen der extrazellulären und der intrazellulären Umgebung der Zelle übertragen.
Im Gegensatz dazu enthalten viele kanalbildende Membranproteine, wie z. B. Porine, mehrere Beta-Stränge des Proteins. Diese bilden Wasserstoffbrückenbindungen, um ein kontinuierliches zylindrisches Beta-Faltblatt zu bilden, wodurch eine starre ringförmige Struktur entsteht, die als Beta-Fass bezeichnet wird.
Die Aminosäuren in den Strängen wechseln zwischen polaren und unpolaren Resten. Unpolare Gruppen zeigen nach außen und interagieren mit der hydrophoben Membran.
Die polaren Seitenketten orientieren sich an der inneren hydrophilen Öffnung, die einen Kanal vom extrazellulären zum intrazellulären Raum bildet, der den Durchgang kleiner polarer gelöster Stoffe ermöglicht.

Bei Multi-Pass-Transmembranproteinen durchquert die Polypeptidkette die Membran mehr als einmal. Die Transmembran-Polypeptidkette bildet entweder eine α-Helix- oder eine β-Strang-Struktur. α-Helix-haltige Multi-Pass-Transmembranproteine sind allgegenwärtig, wohingegen β-Strang-haltige Proteine hauptsächlich in gramnegativen Bakterien, Mitochondrien und Chloroplasten vorkommen.

α-Helix mit Multi-Pass-Transmembranproteinen

Multi-Pass-Transmembranproteine wie G-Protein-gebundene Rezeptoren (GPCRs) und Bakteriorhodopsin enthalten mehrere Transmembrandomänen. Alle GPCRs haben sieben Transmembran-α-Helices, aber jeder Rezeptor hat seine spezifische extrazelluläre Domäne und G-Protein-Bindungsstelle. GPCRs binden einen Liganden und aktivieren ein Membranprotein namens G-Protein. Das aktivierte G-Protein interagiert dann entweder mit einem Ionenkanal oder einem Enzym in der Membran.

Bakteriorhodopsin, das in bestimmten photosynthetischen Bakterien vorkommt, verfügt über sieben Transmembran-α-Helices. Es handelt sich um eine lichtbetriebene Protonenpumpe, die die ATP-Produktion unterstützt, indem sie einen Protonengradienten über die Membran erzeugt.

β-Stränge, die Multi-Pass-Transmembranproteine enthalten

Einige bakterielle Membranproteine enthalten transmembrane β-Stränge, die in einer ringförmigen Struktur namens β-Fass angeordnet sind. Zur Bildung der fassartigen Struktur sind mindestens acht β-Stränge erforderlich, wobei die Kanten der beiden eng beieinander liegenden β-Stränge miteinander Wasserstoffbrückenbindungen bilden. Das in der Membran einiger gramnegativer Bakterien beobachtete Porinprotein enthält ein transmembranöses β-Fass. Neben Bakterien kommen Porinproteine auch in Mitochondrien- und Chloroplastenmembranen vor, was die Hypothese stützt, dass sowohl Mitochondrien als auch Chloroplasten aus Prokaryoten hervorgegangen sind.

Membrane-spanning proteins can cross the cell membrane more than once. Their transmembrane chain usually folds into either an alpha-helix or a beta-strand. Alpha-helix multi-pass proteins are common, while beta-strand proteins are found mainly in gram-negative bacteria, mitochondria, and chloroplasts.

Some multi-pass alpha-helix proteins are G-protein-linked receptors, or GPCRs, and bacteriorhodopsin. GPCRs have seven transmembrane alpha-helices. Each receptor also has its own extracellular domain and G-protein-binding site. They bind a ligand and activate a membrane protein called a G-protein, which then interacts with either an ion channel or an enzyme in the membrane.

Bacteriorhodopsin is found in certain photosynthetic bacteria. It also has seven transmembrane alpha-helices. This protein acts as a light-driven proton pump. By building a proton gradient across the membrane, it helps drive ATP production.

Some bacterial membrane proteins use transmembrane beta-strands instead of helices. These beta-strands arrange into a ring-shaped structure called a beta-barrel. At least eight beta-strands are needed to form this barrel-like shape. The edges of two closely spaced beta-strands form hydrogen bonds with each other.

Porin is a well-known beta-barrel protein found in the membranes of some gram-negative bacteria. Porin proteins are also found in mitochondrial and chloroplast membranes. This distribution supports the idea that mitochondria and chloroplasts evolved from prokaryotes.
