Mechanisms of Membrane Domain Formation

Membrandomänen bilden sich, wenn spezifische Protein-Protein-, Protein-Lipid- und Lipid-Lipid-Wechselwirkungen unterschiedliche Bereiche innerhalb der Membran erzeugen.

Einige Proteine haben eine hohe Affinität zu bestimmten Lipiden, wodurch größere lipidinduzierte Domänen entstehen. Diese Domänen konzentrieren Proteine und ermöglichen die Bildung funktioneller Proteinkomplexe.

Einige Membrandomänen werden aufgrund von Protein-Protein-Wechselwirkungen gebildet, während andere aufgrund der Anwesenheit spezifischer Lipide wie Sphingolipide und Sterole gebildet werden – beispielsweise aggregieren große Proteine wie Bakteriorhodopsin und bilden unterschiedliche Domänen.

Ein weiterer Mechanismus für die Bildung von Membrandomänen beinhaltet die Interaktion von Membranproteinen mit Zytoskelett- oder Exoplasmaproteinen. Beispielsweise interagiert Spectrin, das in roten Blutkörperchen vorkommende Zytoskelettprotein, direkt mit integralen Membranproteinen und immobilisiert diese. Die innere Schicht einer Plasmamembran, die eng mit dem Protein-Zytoskelett-Netzwerk verbunden ist, bildet unterschiedliche Kompartimente oder Korrale. Die Bewegung der in den Ställen vorhandenen Proteine wird oft durch den Zaun der Ställe eingeschränkt. Solche Einschränkungen verhindern die laterale Diffusion der Proteine und führen zur Bildung der Proteindomänen.

Proteine, die an Zell-Zell-Interaktionen beteiligt sind, binden an Proteine benachbarter Zellen. Diese Wechselwirkungen zwischen den beiden Zelloberflächen fixieren die Membranproteine und führen zur Bildung von Proteindomänen. Tight Junctions sind beispielsweise Membranmikrodomänen, die eine wasserdichte Abdichtung zwischen zwei benachbarten tierischen Zellen bilden. Vorwiegend zwei Proteine, Claudine und Occludine, halten die Zellen fest aneinander. Darüber hinaus sind Desmosomen in tierischen Zellen weitere hochgeordnete Membrandomänen, die wie Schweißpunkte zwischen benachbarten Epithelzellen wirken. Cadherine, kurze Proteine in der Plasmamembran, verbinden sich über Linkerproteine wie Desmoplakin, Plakoglobin und Plakophilin mit Zwischenfilamenten, um Desmosomen zu bilden. Die Cadherine verbinden zwei benachbarte Zellen und halten die Zellen in dehnbaren Organen und Geweben wie Haut, Herz und Muskeln in einer blattartigen Formation.

Cell membranes form domains when lipids and proteins gather into distinct regions. These regions, or membrane domains, are shaped by different physical properties that help some molecules stay together while others move apart.

Some domains form through protein-protein interactions. Others depend on specific lipids, such as sphingolipids and sterols. Large proteins, including bacteriorhodopsin, can also cluster and create visible membrane domains.

Membrane proteins can be held in place by cytoskeletal or exoplasmic proteins. In red blood cells, spectrin, a cytoskeletal protein, directly interacts with integral membrane proteins and immobilizes them. The membrane region linked to the protein cytoskeleton can form compartments, or corrals, where protein movement is restricted by a fence-like boundary. This limited lateral diffusion helps produce protein domains.

Cell-cell interactions can also lock membrane proteins into place. Tight junctions are membrane microdomains that form a watertight seal between adjacent animal cells. Claudins and occludins are the main proteins that hold these cells tightly together.

Desmosomes are another highly ordered type of membrane domain in animal cells. They act like spot welds between adjacent epithelial cells. Cadherins in the plasma membrane connect to intermediate filaments through linker proteins such as desmoplakin, plakoglobin, and plakophilin. These connections help keep cells in a sheet-like arrangement in tissues that stretch, including the skin, heart, and muscles.
