Carbonylverbindungen, die Wasserstoff an der chiralen α Kohlenstoff enthalten, werden, wenn sie mit Säuren oder Basen behandelt werden, durch Bildung eines achiralen Enols racemisiert.
Im sauren Medium entsteht durch die Protonierung des C=O-Sauerstoffs und die anschließende Deprotonierung des α-Kohlenstoffatoms ein Enol.
Im basischen Medium führt die Deprotonierung des α-Kohlenstoffatoms zur Bildung eines resonanzstabilisierten achiralen Enolat-Zwischenprodukts.
Anschließend entsteht durch Protonierung des Enolatsauerstoffs das Enol.
In beiden Fällen kann die Reprotonierung von beiden Seiten der Ebene der Doppelbindung mit gleicher Wahrscheinlichkeit auftreten, da das gebildete Enol achiral ist.
Dies führt zur Bildung eines racemischen Gemischs – eines mit zurückgehaltener und das andere mit invertierter Stereochemie.
Falls die Carbonylverbindung einen zusätzlichen chiralen Kohlenstoff trägt, verändert die Enolisierung die Stereochemie des α Kohlenstoffs, um ein stabileres Isomer zu erzeugen.

Unter saurem Medium wird ein Sauerstoffatom der Carbonylgruppe unter gleichzeitiger Entfernung des α-Wasserstoffs protoniert, wodurch ein Enol entsteht.

Alternativ wird im basischen Medium durch die Entfernung von α-Wasserstoff ein resonanzstabilisiertes Enolat erzeugt. Anschließend ergibt die Protonierung des Enolatsauerstoffs ein Enol. Es ist zu beachten, dass die Deprotonierung einer Carbonylverbindung zur Bildung von Enol begünstigt ist, wenn die C(α)-H(α)- und C=O-Bindungen senkrecht zueinander stehen, was die Überlappung von σ-Orbital und π-Orbital ermöglicht.

Da das Enolalken eine planare Geometrie aufweist, kann das eintreffende elektrophile Proton das Molekül entweder von oben oder von unten mit der gleichen Wahrscheinlichkeit angreifen, was zu einer racemischen Mischung von Enantiomeren führt. In dieser racemischen Mischung bleibt die Stereochemie eines der Enantiomere erhalten, während sie für das andere invertiert ist.

Racemization can happen when a carbonyl compound forms an enol. In these molecules, the chiral alpha-carbon is the stereocenter. When the molecule changes under acidic or basic conditions, the original chiral center can be lost.

In acidic solution, the carbonyl oxygen is protonated first. At the same time, the alpha-hydrogen is removed, and an enol forms. In basic solution, removal of the alpha-hydrogen gives a resonance-stabilized enolate. The enolate oxygen is then protonated to make an enol.

The change to an enol is favored when the C(alpha)–H(alpha) bond and the C=O bond are perpendicular. This arrangement allows overlap between a sigma orbital and a pi orbital. The enol alkene is planar, so an incoming electrophilic proton can attack from either the top or the bottom of the plane with equal chance.

Because either face can be attacked, a racemic mixture of enantiomers forms. In this mixture, one enantiomer keeps its stereochemistry, while the other is inverted. This is how enolization can erase and then remake stereochemical information at the alpha-carbon.
