Zellverbindungen sind große Multiproteinkomplexe, die Zellen mit der extrazellulären Matrix verbinden und Zell-Matrix-Verbindungen bilden können, oder als Kontaktpunkte zwischen benachbarten Zellen in Geweben fungieren können, um Zell-Zell-Verbindungen zu bilden.
Zellen haben typischerweise drei Haupttypen von Zell-Zell-Verbindungen - enge, Verankerungs- und Gap-Verbindungen, die jeweils eine unterschiedliche Struktur und Funktion aufweisen.
Betrachten wir das säulenförmige Epithel. Die Tight Junctions in der Nähe der Spitzen dieser Zellen halten benachbarte Plasmamembranen zusammen und fungieren als Barriere für den Fluss von Molekülen durch die Interzellularräume.
Unterhalb dieses apikalen Abschnitts gibt es zwei Arten von Verankerungsübergängen – die Adhären-Verbindungen und die Desmosomen.
Zusammen bilden diese Verankerungsverbindungen ein miteinander verbundenes Zellskelett-Netzwerk, das hilft, mechanischen Kräften zu widerstehen.
Der dritte Typ, die Gap Junctions, sind in der gesamten Plasmamembran verteilt, insbesondere in Richtung des basalen Endes. Diese Verbindungen fungieren als Kommunikationskanäle zwischen benachbarten Zellen und ermöglichen den Durchgang von Molekülen und Ionen.

Overview of Cell-Cell Junctions

Die komplexe dreidimensionale Anordnung der Zellen in jedem mehrzelligen Organismus wird durch die Wechselwirkungen der Zellen untereinander und mit der extrazellulären Matrix definiert und aufrechterhalten. Zell-Zell-Verbindungen sind spezialisierte Strukturen, bei denen die Multiproteinkomplexe einer Zelle mit den Multiproteinkomplexen einer anderen Zelle interagieren. Diese Zellverbindungen werden anhand ihrer Funktion in drei Haupttypen eingeteilt: Verschluss-, Verankerungs- und Gap-Junctions.

Verschließende oder enge Verbindungen

Tight Junctions dienen in erster Linie dazu, die Bewegung von Molekülen wie Wasser und gelösten Substanzen  durch die Interzellularräume zu verhindern. Sie liegen als eine Reihe von Transmembranproteinen vor, die direkt mit denselben Proteinen auf der benachbarten Membran interagieren. Dadurch halten sie die beiden Plasmamembranen in unmittelbarer Nähe zueinander und verhindern so den Durchgang von Molekülen. Tight Junctions können auch selektiv durchlässig sein, sodass ladungsspezifische Ionen durchgelassen werden können, während andere blockiert werden.

Verankerungsknotenpunkte

Verankerungsverbindungen halten Zellen zusammen und verleihen der Gewebestruktur mechanische Festigkeit. Sie bilden Kontaktpunkte, an denen das Zytoskelett einer Zelle über transmembranäre Adhäsionsmoleküle, sogenannte Cadherine, mit dem der Nachbarzellen verankert ist. Es gibt zwei Arten von Ankerverbindungen:

	<li>adhärente Verbindungen, die das Aktin-Zytoskelett und verbinden</li>
	<li>Desmosomen, die die Zwischenfilamente verbinden.</li>
</ol>

Somit bilden sie ein umfangreiches Zytoskelettnetzwerk, das kooperativ zur Aufrechterhaltung der Gewebestruktur und -funktion beiträgt.

Gap Junctions

Gap Junctions sind Proteinkanäle, die die Kommunikation zwischen Zellen erleichtern. Die beteiligten Transmembranproteine bilden einen Kanal, der die Zytoplasmen benachbarter Zellen verbindet und den Austausch von Molekülen und elektrischen Impulsen ermöglicht. Die Bewegung der Moleküle wird durch das Öffnen und Schließen der Kanalproteine reguliert. Beispielsweise wird das Signal für rhythmische Kontraktionen des Herzens über die Gap Junctions im Herzmuskelgewebe von Zelle zu Zelle weitergeleitet. Aufgrund der Zellwand können Pflanzenzellen jedoch keine Moleküle über Gap Junctions austauschen. Stattdessen bilden sie Kommunikationskanäle, sogenannte Plasmodesmen. Hierbei handelt es sich um mit Membranen ausgekleidete Poren, die die Zellen durch die Zellwände verbinden, sodass das Zytoplasma einer Zelle an das einer anderen Zelle angrenzt. Somit besteht eine freie Bewegung gelöster Substanzenund Ionen durch Pflanzenzellen, die durch Plasmodesmen verbunden sind.

Cell junctions help hold tissues together and let neighboring cells communicate. In multicellular organisms, the three-dimensional shape of tissues depends on interactions between cells and the extracellular matrix. Cell-cell junctions are specialized structures made of multi-protein complexes on one cell that connect with similar complexes on another cell.

These junctions are grouped into three main types based on function: occluding, anchoring, and gap junctions. Tight junctions, also called occluding junctions, stop water and solutes from moving through the spaces between cells. They do this with transmembrane proteins that match and bind to the same proteins on the adjacent cell membrane, keeping the two membranes close together. Tight junctions can also be selectively permeable, so some charge-specific ions can pass while others are blocked.

Anchoring junctions give tissues mechanical strength by holding cells together. They create contact points where the cytoskeleton of one cell attaches to the cytoskeleton of a neighboring cell through transmembrane adhesion molecules called cadherins. Two main forms are adherens junctions, which link actin filaments, and desmosomes, which link intermediate filaments. Together, these connections build a strong cytoskeletal network that supports tissue structure and function.

Gap junctions are channels that let cells communicate with each other. Their transmembrane proteins form a passage between the cytoplasms of adjacent cells, allowing molecules and electrical impulses to move across. The channels can open and close to control movement. In cardiac muscle, gap junctions help pass the signal for rhythmic heart contractions from cell to cell.

Plant cells do not use gap junctions because of the cell wall. Instead, they connect through plasmodesmata, which are membrane-lined pores that pass through the cell walls. These channels link the cytoplasm of one plant cell to another, allowing solutes and ions to move freely between them.
