Cadherine sind eine Superfamilie von Calcium-abhängigen Zelladhäsionsmolekülen oder CAMs.
Alle Cadherine haben eine Grundstruktur, die aus einer kleinen C-terminalen zytosolischen Domäne, einer einzelnen Transmembrandomäne und mehreren extrazellulären Cadherin- oder EC-Domänen besteht.
Diese EC-Domänen sind durch flexible Scharnierbereiche miteinander verbunden, die jeweils mit drei Kalzium-Bindungstaschen ausgestattet sind.
Wenn Kalziumionen binden, wird das Cadherin starr, was eine cis- und trans-Bindung an andere Cadherine ermöglicht.
Bei der cis-Bindung können Cadherine auf derselben Zelle aneinander binden und Cluster bilden, während die Interaktion der EC-Domänen von Cadherinen auf benachbarten Zellen zu einer Trans-Bindung führt.
Die N-terminale EC-Domäne, die am weitesten von der Membran entfernt ist, EC1 genannt, hat eine Knopf- und Taschenstruktur. Bei der Trans-Bindung passt der Knopf einer EC1 in die Tasche einer anderen EC1 auf der benachbarten Zelle.
Dies führt dazu, dass Cadherine eines Typs bevorzugt an Cadherine des gleichen Typs binden, eine homophile Bindung.
Obwohl die Wechselwirkung zwischen einem einzelnen Paar von Cadherinen relativ schwach ist, können die Cadherin-Cluster eine kollektiv starke trans-Bindung bilden. Solche Komplexe bilden verankernde Zellverbindungen.

Die Cadherine waren eines der ersten entdeckten Zelladhäsionsmoleküle; Der Begriff „Cadherine“ basiert auf ihren kalziumabhängigen Adhäsionseigenschaften. Die ersten Cadherine, die auf Epithel-, Neuronen- und Plazentazellen entdeckt wurden, wurden E-Cadherin, P-Cadherin bzw. N-Cadherin genannt. Diese klassischen Cadherine weisen Sequenz- und Strukturähnlichkeiten auf. Andere Cadherine, einschließlich derjenigen, die an der Zellsignalisierung beteiligt sind, werden in nichtklassische Cadherine eingeteilt. Diese Vielfalt an Cadherinen entsteht durch mehrere Cadherin-Gene im Genom und alternatives RNA-Spleißen während der Transkription.

Cadherine in Zellverbindungen

Cadherine sind Transmembranproteine mit mehreren extrazellulären Bindungsdomänen. Ihre zytosolische Domäne bindet Linkerproteine, die eine Verbindung zum Zytoskelett herstellen. Cadherine gruppieren sich durch cis-Bindung auf der Plasmamembran und bilden große Multiproteinkomplexe, sogenannte Zellverbindungen. Während klassische Cadherine wie E-Cadherin adhärente Verbindungen bilden, bilden nichtklassische Cadherine wie Desmoglein und Desmocollin desmosomale Verbindungen. Diese Zellverbindungen dienen dazu, die Zellen zusammenzuhalten und Geweben wie dem Epithel mechanische Festigkeit zu verleihen.

Klinische Bedeutung von Cadherinen

Cadherine spielen eine wesentliche Rolle bei der Zelladhäsion, der Gewebebildung und der interzellulären Kommunikation. Folglich kann sich jede Veränderung ihrer Struktur, Ausprägung und Stabilität in klinischen Zuständen manifestieren. Wenn die Zellen den Kontakt zu benachbarten Zellen verlieren, können sie sich aus dem Gewebe lösen und in andere Gewebe eindringen. Der Verlust der Zelladhäsion aufgrund von Mutationen in Cadherin-Genen ist mit der Bösartigkeit von Tumoren verbunden, wie sie beispielsweise bei bösartigen Brustzellen auftreten. Krankheitserreger nutzen Cadherine auch als Rezeptoren, um sich an die Wirtszelle anzuheften und in diese einzudringen. Beispielsweise wird das Cadherin CDHR3 stark im Lungenepithel exprimiert und vom Rhinovirus der Klasse C (RV-C), das Erkältungen verursacht, als Rezeptor verwendet.

Cadherins are cell adhesion proteins that help cells stick together and build tissues. Their name comes from their calcium-dependent binding. Early examples were named E-cadherin, P-cadherin, and N-cadherin after the epithelial, placental, and neuronal cells where they were first found.

Classical cadherins share similar sequences and structures. Other cadherins, including those involved in cell signaling, are grouped as non-classical cadherins. This diversity comes from multiple cadherin genes in the genome and from alternative RNA splicing during transcription.

Cadherins are transmembrane proteins. They have several extracellular binding domains and a cytosolic domain inside the cell. The cytosolic region binds linker proteins, which connect cadherins to the cytoskeleton. Cadherins also cluster on the plasma membrane through cis-binding, which helps form large protein complexes called cell junctions.

Classical cadherins such as E-cadherin form adherens junctions. Non-classical cadherins such as desmoglein and desmocollin form desmosomal junctions. These junctions hold cells together and give mechanical strength to tissues such as epithelium.

Cadherins are important for cell adhesion, tissue formation, and communication between cells. Changes in their structure, expression, or stability can lead to disease. When cells lose contact with neighboring cells, they may break free from the tissue and invade other tissues. Mutations in cadherin genes are linked to tumor malignancy, including in breast cancer cells. Some pathogens also use cadherins as receptors to attach to and enter host cells. One example is CDHR3, which is highly expressed in lung epithelium and is used by class-C rhinovirus, or RV-C, the virus that causes the common cold.
