Catenine sind eine vielfältige Gruppe von Adapterproteinen, die an der Zell-Zell-Adhäsion beteiligt sind.
Catenine haben mehrere Bindungsdomänen, die Cadherine, Zytoskelettkomponenten und verschiedene Zytoskelett-Regulatorproteine binden. So verknüpfen sie das Zytoskelett mit Cadherinen an den Adhäsionsstellen der Zellen.
Während einige Catenine, wie z. B. Plakoglobin und Plakophilin, dazu beitragen, stabile Verbindungen zwischen Cadherinen und den Zwischenfilamenten herzustellen, organisieren andere, wie Alpha- und Beta-Catenine, das Aktin-Zytoskelett dynamisch an der Plasmamembran.
Die Organisation der Aktinfilamente beginnt mit der Bindung von Beta-Catenin an die zytosolische Domäne eines Cadherinmoleküls. Anschließend bindet Alpha-Catenin an das Beta-Catenin.
Ein weiteres Catenin, p120, bindet ebenfalls an diese Komplexe und reguliert die Stabilität der Cadherine auf der Membran.
Wenn sich einige wenige Cadherin-Catenin-Komplexe zusammenschließen, dissoziieren die Alpha-Catenine und bilden zytosolische Dimere.
Diese Alpha-Catenin-Dimere interagieren stark mit dem filamentösen Aktin und helfen bei der Organisation von Aktinbündeln, wodurch die Zelladhäsionsstellen mit Zytoskelettstärke versorgt werden.

Catenins

Catenine zeichnen sich durch mehrere Bindungsdomänen und dynamische Strukturen aus, die es ihnen ermöglichen, als Linkerproteine in Zellverbindungskomplexen zu fungieren. Alle Catenine, mit Ausnahme von α-Catenin, enthalten eine charakteristische Proteinsequenz, die Armadillo-Repeat genannt wird, und werden daher auch Armadillo-Proteine genannt.

Catenine in Zellverbindungen

Catenine binden an Zelladhäsionsmoleküle wie Cadherine und verknüpfen diese je nach Art der Zellverbindung mit unterschiedlichen Proteinen des Zytoskeletts. An der Adherens-Verbindung wird das Aktin-Zytoskelett direkt oder indirekt dynamisch durch die Alpha-, Beta- und p120-Catenine organisiert. Beispielsweise bindet α-Catenin direkt an F-Aktin, während p120 die Aktinpolymerisation über Rho-GTPasen reguliert. Bei Desmosomen werden die Zwischenfilamente jedoch durch die Wechselwirkungen spezieller Catenine namens Plakoglobin und Plakophilin mit anderen Proteinen wie Desmoplakin organisiert.

β-Catenin und Zellsignalisierung

Zusätzlich zur Bindung von Cadherinen an Zellverbindungen kann β-Catenin auch bei der Zellsignalisierung wie dem Wnt-Signalweg eine Rolle spielen. Auf diesem Weg löst die Bindung eines Wnt-Signalmoleküls an den Zelloberflächenrezeptor, der als Frizzled bezeichnet wird, eine Kaskade aus, die die β-Catenin-Konzentration im Zytoplasma erhöht. Dadurch kann β-Catenin in den Zellkern wandern, an Transkriptionsfaktoren binden und die Zielgenexpression regulieren. Im Gegensatz zu anderen Cateninen, die in verschiedenen Organismen mehrere gewebespezifische Varianten aufweisen, kommt bei Wirbeltieren nur eine einzige Variante von β-Catenin vor. Diese Variante besteht aus einer hochkonservierten Aminosäuresequenz bei allen Wirbeltierarten.

p120-Catenin und synaptische Plastizität

Die Cadherin-Catenin-Komplexe sind ein wichtiger Teil der synaptischen Plastizität – der Stärkung und Schwächung synaptischer Verbindungen, die am Gedächtnis beteiligt sind. Beispielsweise ist p120 an der Stabilisierung von Glutamatrezeptoren auf der synaptischen Membran beteiligt und sorgt so für eine langfristige Potenzierung. Es ist auch an der Stabilisierung neuronaler Cadherine beteiligt, indem es deren Endozytose verhindert und die Cadherin-Clusterbildung an synaptischen Verbindungen fördert. Es hat sich gezeigt, dass die Herunterregulierung von p120 die Adhäsion zwischen den prä- und postsynaptischen Neuronen verhindert.

Catenins help cells stay connected and also support signaling inside the body. These proteins act as linker proteins in cell junction complexes because they have multiple binding domains and flexible structures. Most catenins contain a protein sequence called the armadillo repeat, so they are also known as armadillo proteins. Alpha-catenin is the main exception.

Catenins bind to cell adhesion molecules such as cadherins. They then connect cadherins to cytoskeletal proteins, and the exact link depends on the type of cell junction. At adherens junctions, alpha-catenin, beta-catenin, and p120-catenin help organize the actin cytoskeleton. Alpha-catenin binds directly to F-actin. p120 helps control actin polymerization through Rho GTPases.

At desmosomes, catenins help organize intermediate filaments instead of actin. In this case, special catenins called plakoglobin and plakophilin interact with other proteins such as desmoplakin. These interactions help build strong cell connections in tissues that need mechanical support.

Beta-catenin also has a signaling role in the Wnt pathway. When a Wnt signal molecule binds to the frizzled receptor on the cell surface, a cascade raises beta-catenin levels in the cytoplasm. Beta-catenin can then move into the nucleus, bind to transcription factors, and regulate target gene expression. In vertebrates, only one beta-catenin variant is found, and its amino acid sequence is highly conserved across species.

p120-catenin also plays an important role in synaptic plasticity, which is the strengthening and weakening of synaptic connections involved in memory. It helps stabilize glutamate receptors on the synaptic membrane and supports long-term potentiation. p120 also stabilizes neural cadherins by preventing their endocytosis and by promoting cadherin clustering at synaptic junctions. When p120 is downregulated, adhesion between pre- and postsynaptic neurons is reduced.
