Desmosomen sind Verankerungsverbindungen, die häufiger in Geweben vorkommen, die mechanischer Belastung ausgesetzt sind, wie z. B. Herzmuskel und Darmepithel.
Die Zelladhäsion an Desmosomen wird durch desmosomale Cadherine, nämlich Desmocollin und Desmoglein, erleichtert, die über heterophile Wechselwirkungen ihrer extrazellulären Domänen adhäsieren.
Die zytoplasmatische Domäne der Cadherine bindet Catenine, die als Plakoglobine und Plakophiline bezeichnet werden und auf der zytoplasmatischen Seite eine dichte Plaque bilden.
Die Catenine können nicht direkt an intermediäre Filamente binden und sind mit ihnen durch Desmoplakin - ein zytoskelettales Linkerprotein aus der Familie der Plakin - verbunden.
Die N-terminale Domäne von Desmoplakin bindet die Catenine, und ihre C-terminalen Plakin-Wiederholungen binden die Zwischenfilamente.
Auf diese Weise verankert das Desmosom die intermediären Filamente benachbarter Zellen und bildet ein transzelluläres Zytoskelett-Netzwerk über das Gewebe.

Desmosomes

Die Cadherine sind eine Superfamilie von Zelladhäsionsmolekülen mit über 180 Varianten, wobei bestimmte Gewebe eine bestimmte Kombination von Cadherin-Typen exprimieren. Cadherine weisen im Allgemeinen eine homophile Bindung auf; Das heißt, Cadherine auf einer Zelle binden an Cadherine desselben oder eines eng verwandten Typs auf einer anderen Zelle. Daher haben Zellen desselben Typs eine spezifische Affinität, sich aneinander zu binden und sich in Clustern zu ordnen, um Gewebe zu bilden.

Zellsortierung während der Entwicklung

Die Zellsortierung spielt während der Embryogenese und Entwicklung eine wesentliche Rolle, wie frühe Experimente mit Amphibienembryonen gezeigt haben. Zellen von jungen Embryonen wurden dissoziiert und man ließ sie vermischen. Als man diese Mischung aggregieren ließ, gruppierten sich ähnliche Zellen und bildeten Strukturen. Diese Strukturen ähnelten embryonalen Schichten wie dem Mesoderm, der Epidermis und dem Neuralrohr. Die Cadherin-Expression ändert sich, wenn sich die Zellen während der Entwicklung differenzieren, was die Bildung komplexer Strukturen wie des Neuralrohrs von Wirbeltieren ermöglicht. Während sich die Nervenfurche aus der ektodermalen Schicht bildet, verlagern die Neuralplattenzellen ihre Cadherin-Expression von E-Cadherin zu N-Cadherin. Die Zellen am Rand der Neuralplatte beginnen auch, andere Cadherine wie Cadherin 6B zu exprimieren, wodurch die beiden Enden zusammengeführt werden und das Abklemmen des Neuralrohrs erleichtert wird.

Cadherine und Malignität

Die durch Cadherin vermittelte Zell-Zell-Adhäsion ist ein wichtiger Mechanismus, der die Gewebehomöostase in mehrzelligen Organismen aufrechterhält. In Krebsgewebe bricht diese Zelladhäsion jedoch zusammen. Viele epitheliale Tumorzellen stellen die Expression von E-Cadherin-Molekülen teilweise oder vollständig ein, unterstützt durch verschiedene Transkriptionsregulatoren. Beispielsweise hemmt ein Transkriptionsregulator namens Twist die Expression von E-Cadherin und verhindert so die Anheftung der Tumorzellen an die Nachbarzellen. Diese Transformation wird als epithelialer zu mesenchymaler Übergang (EMT) bezeichnet. Es ist ein charakteristisches Merkmal des Fortschreitens zur Malignität, das es Tumorzellen ermöglicht, zu entweichen und in andere Gewebe einzudringen. Die experimentelle Blockierung von Twist in bösartigen Brustkrebszellen verringert deren Bösartigkeit. Im Gegensatz dazu können normale Epithelzellen dazu veranlasst werden, sich einer EMT zu unterziehen, indem die Twist-Expression erhöht wird.Desmosomen

Der Begriff Desmosom leitet sich von den griechischen Wörtern „desmo“ und „soma“ ab, was „Haftkörper“ bedeutet. Diese Struktur wurde erstmals im späten 19. Jahrhundert beobachtet und als kleine, dichte Knötchen in der Epidermis beschrieben. Desmosomen sind knopfartige Strukturen, die dabei helfen, ein miteinander verbundenes Netzwerk aus Zwischenfilamenten über die Zellen hinweg zu bilden. Diese Verbindungen sind wichtig, um Zellen unter mechanischer Belastung zusammenzuhalten und die Gewebeintegrität aufrechtzuerhalten. Desmosomen sind Multiproteinkomplexe aus desmosomalen Cadherinen, die über verschiedene Adapterproteine mit Zwischenfilamenten verbunden sind.

Desmosomale Cadherine

Die Cadherine bilden die extrazelluläre Kernregion eines Desmosoms und binden Cadherine auf der angrenzenden Zelle. Es gibt zwei Arten von Cadherinen in Desmosomen – Desmogleine und Desmocolline. Diese Transmembran-Glykoproteine besitzen einen großen extrazellulären Anteil, der aus vier Cadherin-Wiederholungen und einer extrazellulären Ankerdomäne besteht, die diese Proteine an der Zellmembran verankert. Heterodimere Paare von Desmogleinen und Desmocollinen bilden Cluster durch cis-Wechselwirkungen auf derselben Zelle und trans-Wechselwirkungen zwischen benachbarten Zellen. Ihre zytoplasmatischen Domänen können aufgrund des alternativen Spleißens zwischen den verschiedenen Isoformen in der Länge variieren – zum Beispiel gibt es beim Menschen vier Desmoglein-Isoformen und drei Desmocollin-Isoformen.

Adapterproteine in Desmosomen

Die zytoplasmatischen Domänen desmosomaler Cadherine binden die Gürteltierfamilie der Proteine Plakoglobin und Plakophilin, die eine dichte Plaque bilden. Diese Proteine verfügen über die charakteristischen Arm-Repeats, die als Andockstelle für verschiedene Linkerproteine wie Desmoplakin fungieren. Desmoplakin ist das am häufigsten vorkommende Protein in einem desmosomalen Komplex. Ihr Carboxyterminal enthält die Plakin-Wiederholungen, die das Zwischenfilament binden und so eine Verbindung zwischen der desmosomalen Plaque und dem Zytoskelett bilden.

Erkrankungen der Desmosom-Dysfunktion

Eine desmosomale Dysfunktion äußert sich in der Regel in Störungen des Herzens und der Haut – Gewebe, die typischerweise einer hohen mechanischen Belastung ausgesetzt sind. Beispielsweise können Mutationen in den Desmoglein-Genen eine epidermale Verdickung und eine arrhythmogene rechtsventrikuläre Kardiomyopathie (ARVC) verursachen. Darüber hinaus verursachen Autoantikörper gegen die desmosomalen Cadherine die Autoimmunerkrankung Pemphigus vulgaris, bei der Patienten aufgrund einer schwachen Zell-Zell-Adhäsion epidermale Blasen aufweisen.

Desmosomes are cell junctions that anchor cells together. They are button-like structures that create an interlinked network of intermediate filaments across cells. This strong link helps tissues resist mechanical stress and maintain tissue integrity.

Desmosomes are built from multiple proteins. Their core includes desmosomal cadherins, which are adhesion proteins that bind to cadherins on neighboring cells. The two desmosomal cadherins are desmogleins and desmocollins. These transmembrane glycoproteins have a large extracellular region with four cadherin repeats and one anchor domain that holds them in the cell membrane.

Desmogleins and desmocollins can pair as heterodimers. They form clusters through cis-interactions on the same cell and trans-interactions between adjacent cells. In humans, there are four desmoglein isoforms and three desmocollin isoforms, and their cytoplasmic domains can differ in length because of alternative splicing.

Inside the cell, the cadherins bind armadillo family proteins called plakoglobin and plakophilin. These proteins form a dense plaque. The plaque then recruits desmoplakin, the most abundant protein in a desmosomal complex. Desmoplakin uses its carboxy-terminal plakin repeats to bind intermediate filaments, which connects the desmosome to the cytoskeleton.

Problems with desmosomes often show up in the heart and skin, where cells face high mechanical strain. Mutations in desmoglein genes can cause epidermal thickening and arrhythmogenic right ventricular cardiomyopathy, or ARVC. Autoantibodies against desmosomal cadherins can also cause pemphigus vulgaris, an autoimmune disease that leads to epidermal blisters because cell-cell adhesion becomes weak.
