Mitosis and Cytokinesis

Mitose ist eine Form der Zellteilung, bei der das genetische Material einer Zelle zwischen zwei Tochterzellen aufgeteilt wird. 
Zunächst kondensiert Nukleinchromatin während der Prophase zu x-förmigen Chromosomen, die aus Schwesterchromatidenpaaren bestehen, die an Zentromerübergängen gebunden sind.
Gleichzeitig wandern die Zentrosomen zu gegenüberliegenden Zellseiten. Dabei beginnen Mikrotubuli-Stäbchen aus jedem zu wachsen, die einen netzartigen Spindelapparat bilden.
Als nächstes löst sich die Kernhülle während der Prometaphase auf und die Proteinstrukturen, die als Kinetochore bezeichnet werden, erscheinen auf beiden Seiten der Zentromere, eine für jedes Chromatin.
Sobald sich diese Kinetochore gebildet haben, befestigen sich ausdehnende innere Mikrotubuli an ihnen, wobei jedes Schwesterchromatid an einen anderen Zellpol gebunden ist.
In der Metaphase ordnet der Spindelapparat die Chromosomen so um, dass sie in einer festen Reihe entlang des Äquators der Zelle ausgerichtet sind.
Mit der Anaphase verkürzen sich die an Kinetochoren fixierten Mikrotubuli und die Schwesterchromatiden werden auseinandergezogen. Diese und andere Mikrotubuli-Dynamiken verlängern die Zelle zusätzlich.
Während der Telophase löst sich der Spindelapparat auf, die Chromatiden lösen sich und die Kernhüllen werden neu gebildet.
Schließlich wird die Zelle durch Zytokinese zytoplasmatisch geteilt, wodurch ein Zellpaar entsteht, das genetisch mit seinem Vorläufer identisch ist.

mitose und zytokinese

Bei Eukaryoten ist der Zellteilungszyklus in verschiedene, koordinierte zelluläre Prozesse unterteilt, zu denen Zellwachstum, DNA-Replikation/Chromosomenverdopplung, Chromosomenverteilung auf Tochterzellen und schließlich Zellteilung gehören. Der Zellzyklus wird durch seine Regulierungssysteme sowie extrazelluläre Signale, die die Zellproliferation beeinflussen, streng reguliert.

Die Prozesse des Zellzyklus laufen über etwa 24 Stunden (in typischen menschlichen Zellen) und in zwei unterscheidbaren Hauptstadien ab. Die erste Stufe ist die DNA-Replikation während der S-Phase der Interphase. Die zweite Stufe ist die mitotische (M) Phase, die die Trennung der duplizierten Chromosomen in zwei neue Kerne (Mitose) und die zytoplasmatische Teilung (Zytokinese) umfasst. Die beiden Phasen sind durch Intervalle (G1- und G2-Lücke) getrennt, in denen sich die Zelle auf die Replikation und Teilung vorbereitet.

Der Prozess der Mitose

Die Mitose kann in fünf verschiedene Stadien unterteilt werden: Prophase, Prometaphase, Metaphase, Anaphase und Telophase. Die Zytokinese, die während der Anaphase oder Telophase (je nach Zelle) beginnt, ist Teil der M-Phase, aber nicht Teil der Mitose.

Prophase

Wenn die Zelle in die Mitose eintritt, beginnen ihre replizierten Chromosomen zu kondensieren und werden mithilfe von Proteinen, die als Kondensine bekannt sind, als fadenförmige Strukturen sichtbar. Der mitotische Spindelapparat beginnt sich zwischen den Zentrosomen zu bilden – die während der S-Phase dupliziert wurden – und zu entgegengesetzten Polen der Zelle zu wandern. Die Spindel besteht aus filamentösen Strukturen, sogenannten Mikrotubuli, die aus Tubulin-Proteinmonomeren bestehen. Spindelmikrotubuli beginnen sich in Richtung der kondensierten Chromosomen auszudehnen. Der Nukleolus, ein Bestandteil des Zellkerns, der Ribosomen produziert, verschwindet, was auf den bevorstehenden Zusammenbruch des Zellkerns hinweist.

Prometaphase

Während der Prometaphase wachsen die Mikrotubulifilamente des Spindelapparats weiter und die Chromosomen verdichten sich. Die Kernhülle zerfällt vollständig und die Chromosomen werden freigesetzt. Einige der Mikrotubuli heften sich an die freigesetzten Chromosomen und binden an eine Proteinstruktur namens Kinetochor, die auf dem Zentromer jedes Paares von Schwesterchromatiden vorhanden ist. Spindelmikrotubuli von entgegengesetzten Polen heften sich an die Kinetochoren und fangen die kondensierten Schwesterchromatidpaare ein. Spindelmikrotubuli, die nicht an Chromosomen haften – polare und astrale Mikrotubuli – helfen dabei, die Spindeln auseinanderzudrücken und die Spindelpole an der Zellmembran zu verankern.

Metaphase

Die Spindelmikrotubuli richten jedes Paar der vollständig kondensierten Schwesterchromatiden entlang des Äquators der Zelle aus – an der Metaphasenplatte. Die Zelle ist nun zur Teilung bereit.

Anaphase

Die Mikrotubuli von gegenüberliegenden Spindelpolen, die an der Kinetochorstruktur befestigt sind, verkürzen und trennen die Schwesterchromatiden am Zentromer. Die Kohäsionsproteine, die die Chromatiden zusammenhalten, zerfallen nun. Die Verkürzung der Kinetochor-Mikrotubuli führt dazu, dass jedes Chromatid des Paars – jetzt Chromosomen genannt – zu einem entgegengesetzten Pol wandert.

Telophase

Sobald die Chromosomen die entgegengesetzten Pole der Zelle erreichen, dekondensieren sie und entrollen sich, um Chromatin zu bilden. Die Spindel-Mikrotubuli-Filamente depolymerisieren zu ihren Tubulinmonomeren, die dann als Zytoskelettelemente in Tochterzellen verwendet werden. Um jeden Chromosomensatz herum bilden sich die Kernhüllen neu.

Zytokinese

Während der Zytokinese in tierischen Zellen bilden Aktinfilamente einen kontraktilen Ring in der Plasmamembran, um eine Spaltfurche zu erzeugen, die die Zelle schließlich in zwei Teile zusammendrückt. In Pflanzenzellen verbinden sich Vesikel aus dem Golgi-Apparat, die Glukose, Enzyme und Strukturproteine transportieren, und bilden an der Stelle der ehemaligen Metaphasenplatte eine neue Zellplatte. Die wachsende Zellplatte verschmilzt mit den Plasmamembranen auf beiden Seiten und bildet schließlich eine neue Zellwand, die die Zelle in zwei Teile teilt.

Die Mitose ist nun abgeschlossen und es entstehen zwei Tochterzellen, die mit der Elternzelle identisch sind. In den meisten menschlichen Zellen macht die Mitose etwa eine Stunde des etwa 24-stündigen Zellzyklus aus.

Cell division in eukaryotes follows a tightly regulated cycle that includes cell growth, DNA replication, chromosome duplication, chromosome distribution to daughter cells, and cell division. Regulatory systems and extracellular signals help control when cells divide. In typical human cells, the cycle takes about 24 hours.

The cycle has two major stages. DNA is copied during the S phase of interphase. The mitotic, or M, phase then separates the duplicated chromosomes into two new nuclei and divides the cytoplasm. G1 and G2 gaps come between these stages and give the cell time to prepare for replication and division.

Mitosis is the part of the M phase that separates the nucleus into two sets of chromosomes. It has five stages: prophase, prometaphase, metaphase, anaphase, and telophase. Cytokinesis, the division of the cytoplasm, starts during anaphase or telophase depending on the cell, but it is not part of mitosis.

During prophase, the replicated chromosomes condense and become visible as threadlike structures with the help of condensins, which are proteins that help package chromosomes. The mitotic spindle begins to form between the centrosomes, which were duplicated in S phase, and the centrosomes move to opposite poles of the cell. The spindle is made of microtubules, which are filaments built from tubulin protein monomers, and these microtubules grow toward the chromosomes. The nucleolus disappears, showing that the nucleus is about to break down.

In prometaphase, the chromosomes finish condensing and the nuclear envelope breaks down completely. This releases the chromosomes into the cell. Some spindle microtubules attach to a protein structure called the kinetochore at the centromere of each sister chromatid pair. Microtubules from opposite poles capture the chromatids, while polar and astral microtubules help push the spindle poles apart and anchor them to the cell membrane.

During metaphase, the spindle microtubules line up each pair of fully condensed sister chromatids at the metaphase plate, or the cell equator. This alignment prepares the cell for division. In anaphase, the kinetochore microtubules shorten, the cohesion proteins holding the chromatids together break down, and the sister chromatids separate at the centromere. Each chromatid then moves to an opposite pole and is now considered a chromosome.

In telophase, the chromosomes reach the poles, decondense, and uncoil into chromatin. The spindle microtubules depolymerize into tubulin monomers, which can be used again as cytoskeletal elements in the daughter cells. New nuclear envelopes then form around each set of chromosomes.

Cytokinesis completes cell division in different ways in animal and plant cells. In animal cells, actin filaments form a contractile ring in the plasma membrane and create a cleavage furrow that pinches the cell in two. In plant cells, vesicles from the Golgi apparatus carrying glucose, enzymes, and structural proteins join to form a new cell plate at the former metaphase plate. The cell plate fuses with the plasma membranes and becomes a new cell wall. Mitosis is then complete, producing two daughter cells identical to the parent cell. In most human cells, this process takes about one hour of the full cell cycle.
