Transcription

Transkription ist der Prozess der Synthese von RNA aus einer DNA-Vorlage.
Auf der Matrizen-DNA baut sich der Transkriptions-Präinitiationskomplex um den Kernpromotor eines Gens an. Der Promotor enthält die TATA-Box und eine Initiator-Sequenz. Die allgemeinen Transkriptionsfaktoren binden an der TATA-Box, während die Initiatorsequenz die Transkriptionsstartstelle enthält.
Sobald die notwendigen Komponenten gebunden sind, wickelt der Präinitiationskomplex ein kurzes Stück der DNA stromaufwärts der Transkriptionsstartstelle ab. Danach werden die allgemeinen Transkriptionsfaktoren vom Strang dissoziiert und die RNA-Polymerase beginnt mit der Produktion eines neuen mRNA-Strangs.
Die Nukleotide werden nacheinander hinzugefügt, und die Synthese der mRNA erfolgt in einer Richtung von fünf Primzahlen bis drei Prime, wobei sie aus dem Matrizenstrang abgelesen wird, mit der Ausnahme, dass die Thymidine durch Uridine ersetzt werden.
Dieser neu erstellte mRNA-Strang stellt eine Kopie der Information im kodierenden Strang dar. Die Synthese wird fortgesetzt, bis eine Terminationssequenz gefunden wird, die die neu hergestellte mRNA freisetzt.

Unter Transkription versteht man die Synthese von RNA aus einer DNA-Sequenz durch RNA-Polymerase. Es ist der erste Schritt bei der Herstellung eines Proteins aus einer Gensequenz. Darüber hinaus sind viele andere Proteine und regulatorische Sequenzen an der korrekten Synthese von Messenger-RNA (mRNA) beteiligt. Die Transkriptionsregulation ist für die Differenzierung verschiedener Zelltypen und häufig für die richtige zelluläre Reaktion auf Umweltsignale verantwortlich.

Durch Transkription können verschiedene Arten von RNA-Molekülen entstehen

Bei Eukaryoten wird die DNA zunächst in Prä-mRNA umgeschrieben, die weiter zu reifer mRNA verarbeitet werden kann, die als Matrize für die Proteinsynthese dient. Bei Prokaryoten kann die RNA-Translation in Polypeptide jedoch beginnen, während die Transkription noch läuft. Durch die Transkription können auch verschiedene Arten von RNA-Molekülen entstehen, die kein Protein kodieren, wie etwa microRNAs (miRNAs), Transfer-RNA (tRNA) und ribosomale RNA (rRNA), die alle die Proteinsynthese beeinflussen.

Die Regulierung der Transkription ist für die Entwicklung von zentraler Bedeutung

Mit wenigen Ausnahmen verfügen alle Zellen im menschlichen Körper über die gleichen genetischen Informationen, doch verschiedene Zelltypen regulieren die Transkription während der Entwicklung unterschiedlich. Insbesondere spielt die Transkriptionsregulation eine zentrale Rolle bei der Zelldifferenzierung – dem Prozess der Produktion spezialisierter Zellen, wie z. B. Muskelzellen, aus weniger spezialisierten Vorläuferzellen. Um die spezialisierten Zellen zu produzieren, müssen einige Gene in den Vorläuferzellen aktiviert und andere deaktiviert werden.

Dieser Prozess der Zelldifferenzierung wird durch DNA-bindende Proteine, sogenannte Transkriptionsfaktoren, gesteuert, die den Grad der Transkription von Genen steuern. Beispielsweise empfangen Zellen in der Ektodermschicht des sich entwickelnden Embryos während der frühen Wirbeltierentwicklung mehrere Induktionssignale von Proteinen wie BMP, WNT und SHH. Diese Signale aktivieren Transkriptionsfaktoren, die eine Vielzahl von Genen ein- oder ausschalten. Auf diese Weise bestimmt die Transkriptionsregulation, ob Ektodermzellen zu Hautzellen oder Zellen des Nervensystems werden.
