Cellular Differentiation

Während der zellulären Differenzierung nehmen unspezialisierte Zellen charakteristische Merkmale wie Form, Größe oder Zusammensetzung von Membranproteinen an, um bestimmte Funktionen im Körper zu erfüllen.
Eine befruchtete Eizelle oder Zygote ist hochgradig proliferativ und totipotent, d.h. sie kann sich differenzieren, um alle Zelltypen zu bilden.
Während die Zygote eine Reihe von Teilungen durchläuft, verlieren die Tochterzellen ihre Totipotenz und differenzieren sich, um spezialisierte Zellen im Embryo zu bilden, wie Muskelzellen, Neuronen, Hautepidermis, Herzzellen und Leberzellen.
Obwohl alle diese Körperzellen das gleiche Genom enthalten, exprimieren sie bestimmte Gene, während sie andere ausschalten. Auf diese Weise können sie die Proteine synthetisieren, die für die Ausführung gewebespezifischer Funktionen erforderlich sind.

Alles beginnt mit einer befruchteten Eizelle, aus der sich eine Vielzahl von Zelltypen entwickelt, wie Nervenzellen, Muskelzellen und Epithelzellen, die den Erwachsenen charakterisieren. Während der Entwicklung und im Erwachsenenalter führt die zelluläre Differenzierung dazu, dass Zellen ihre endgültige Morphologie und Physiologie annehmen. Differenzierung ist der Prozess, durch den undifferenzierte Zellen spezialisiert werden, um bestimmte Funktionen auszuführen. 

 Eine Zygote ist eine totipotente Stammzelle mit sehr hohem Differenzierungspotenzial, die alle Zelltypen bilden und einen ganz neuen mehrzellulären Organismus erzeugen kann. Wenn die Zygote aufeinanderfolgende Zellteilungen durchläuft, entstehen pluripotente Stammzellen (PSCs) mit einem engeren Differenzierungspotenzial. PSCs geben einige adulte Stammzellen ab, die sich weiter vermehren und stärker differenzieren. Wenn sich eine Zelle differenziert, kann sie erhebliche Veränderungen in Größe, Form, Stoffwechselaktivität und Gesamtfunktion erfahren. 

 Ausgehend von der befruchteten Eizelle enthalten alle Zellen im Körper die gleiche DNA, aber jeder Zelltyp „liest„ nur die für seine Funktion relevanten Abschnitte der DNA. Mehrere Faktoren wie Hormone oder Signalmoleküle lösen die Zelldifferenzierung aus und schalten spezifische Gene ein, während sie andere abschalten. Der primäre Mechanismus, durch den Gene „ein-„ oder „aus-„ geschaltet werden, sind Transkriptionsfaktoren. Ein Transkriptionsfaktor ist eine Klasse von Proteinen, die an spezifische Gene auf dem DNA-Molekül binden und entweder ihre Transkription fördern oder hemmen. Die Expression spezifischer Gene vermittelt die Synthese notwendiger Proteine, die für eine bestimmte Zellfunktion erforderlich sind und die zelluläre Differenzierung unterstützen.

Cellular differentiation turns unspecialized cells into specialized cells with specific jobs. This process helps a complex organism, such as a human, develop from a single fertilized egg into a body with many cell types. Nerve cells, muscle cells, and epithelial cells are examples of the final cell types that appear during development.

A zygote is the first cell after fertilization. It is a totipotent stem cell, which means it has very high differentiation potential and can form all cell types and even produce a whole new multicellular organism. As the zygote divides, it gives rise to pluripotent stem cells, or PSCs, which have a more limited ability to differentiate. PSCs then produce a few adult stem cells that keep dividing until they become more terminally differentiated.

As cells differentiate, they can change in size, shape, metabolic activity, and overall function. Even though the body’s cells contain the same DNA, each cell type only reads the parts of the DNA that are relevant to its role. This selective reading helps each cell become suited to its own job.

Hormones and signaling molecules can trigger differentiation. In response, specific genes are turned on while others are turned off. Transcription factors, which are proteins that bind to specific genes on DNA, control this switch by promoting or inhibiting transcription. The expression of the right genes leads to the proteins needed for a particular cell function and supports cellular differentiation.
