Quantitative Analysis

Bei der quantitativen Analyse wird die Menge eines oder mehrerer Bestandteile einer Probe bestimmt, deren Zusammensetzung oft bekannt ist. Ist dies nicht der Fall, wird eine vorläufige qualitative Analyse durchgeführt, um die Bestandteile zu identifizieren.
Bei der quantitativen Analyse wird das Ergebnis aus zwei Messungen geschätzt – der Quantifizierung der Probe und der Messung einer Menge, die proportional zum Analyten in Beziehung steht.
Die quantitative Analyse kann in klassische und instrumentelle Methoden eingeteilt werden.
Die klassischen Methoden bewerten die Masse oder das Volumen des Analyten oder seines Derivats, das durch eine chemische Reaktion mit einem Reagenz erhalten wird. Im Gegensatz dazu untersuchen instrumentelle Methoden die physikalische Natur des Analyten in seiner Wechselwirkung mit elektromagnetischer Strahlung.
Da für die quantitative Analyse unterschiedliche Proben und Instrumente verwendet werden, ist die Kalibrierung der Detektoren erforderlich. Dann wird ein Kalibrierdiagramm mit Küvetten erstellt, die unterschiedliche bekannte Mengen desselben Analyten enthalten, das dann hilft, die unbekannte Analytkonzentration in der Probe zu bestimmen.

Wenn die Zusammensetzung der Probe unbekannt ist, wird zunächst eine qualitative Analyse durchgeführt, um ihre Bestandteile zu identifizieren. Dadurch wird sichergestellt, dass in der quantitativen Phase die richtigen Substanzen gemessen werden.

Bei der quantitativen Analyse werden zwei wichtige Messungen durchgeführt: die Probenmenge und eine Eigenschaft, die proportional zur Menge des Analyten (der zu analysierenden Substanz) ist. Dies bildet die Grundlage der Methode und gewährleistet eine genaue Bestimmung der Konzentration des Analyten.

Quantitative Analysen können in zwei Haupttypen eingeteilt werden: klassische Methoden und instrumentelle Methoden. Bei klassischen Methoden wird die Masse oder das Volumen des Analyten oder seiner chemisch abgeleiteten Form gemessen. Beispielsweise misst die gravimetrische Analyse die Masse eines Niederschlags, der aus einer chemischen Reaktion mit dem Analyten entsteht, um dessen Konzentration zu bestimmen. Eine weitere klassische Methode ist die Titration, bei der das Volumen eines Reaktanten gemessen wird, das für eine vollständige Reaktion mit dem Analyten erforderlich ist.

Instrumentelle Methoden bewerten die Wechselwirkung zwischen dem Analyten und physikalischen Kräften wie elektromagnetischer Strahlung. Beispielsweise wird bei der Spektrophotometrie die Konzentration des Analyten durch Messen der von der Probe absorbierten Lichtmenge bestimmt. Ein weiteres Beispiel ist die Atomabsorptionsspektroskopie, bei der die von den Atomen in der Probe absorbierte Lichtmenge Informationen über die Konzentration des Analyten liefert.

Kalibrierungskurven spielen bei der quantitativen Analyse eine entscheidende Rolle. Detektoren und Instrumente müssen mit bekannten Konzentrationen des Analyten kalibriert werden, die normalerweise in Küvetten gegeben werden. Diese bekannten Werte werden verwendet, um ein Kalibrierungsdiagramm zu erstellen. Dieses wichtige Werkzeug gewährleistet die Genauigkeit der Analyse, indem es hilft, die Konzentration des unbekannten Analyten in der Probe zu bestimmen.

Quantitative analysis measures the amount of a specific substance, called the analyte, in a sample. If the sample composition is unknown, qualitative analysis comes first. That step identifies the components so the correct substances can be measured during the quantitative phase.

This method depends on two key measurements. Scientists measure the sample quantity and also a property that is proportional to the amount of analyte. Together, these measurements help determine the analyte concentration accurately.

Quantitative analysis is usually grouped into classical methods and instrumental methods. Classical methods measure mass or volume, either of the analyte itself or of a chemically derived form. Gravimetric analysis is one example. It measures the mass of a precipitate formed in a chemical reaction with the analyte. Titration is another classical method. It measures the volume of a reactant needed to fully react with the analyte.

Instrumental methods use the analyte’s interaction with physical forces, such as electromagnetic radiation. In spectrophotometry, concentration is found by measuring how much light the sample absorbs. Atomic absorption spectroscopy uses the amount of light absorbed by atoms in the sample to show the analyte concentration.

Calibration curves are a major part of quantitative analysis. Detectors and instruments are calibrated with known concentrations of the analyte, often placed in cuvettes. These known values are used to make a calibration plot. The plot helps scientists find the concentration of the unknown analyte in the sample and improves the accuracy of the analysis.
