Ein Raman-Spektralphotometer besteht aus vier Hauptkomponenten: einer Laserquelle, einem Probenhaltesystem, einem Wellenlängenwähler und einem Detektor.
Die Laserquelle sendet einen fokussierten Strahl monochromatischen Lichts aus, typischerweise im sichtbaren oder nahen Infrarotbereich, von dem ein Teil von Molekülen in der Probe gestreut wird.
Proben können in verschiedenen Formen vorliegen, darunter Flüssigkeit, Lösung, transparenter Feststoff, Pulver, Pellet oder Gas.
Das Streulicht wird gesammelt und durch einen Monochromator geleitet, wobei alle bis auf ausgewählte Einzelwellenlängen ausgeschlossen werden. 
Optische Bandenunterdrückungs- oder "Kerb"-Filter entfernen Licht aus Streulaserstrahlung und Rayleigh-Streuung, die das Raman-Signal stören können.
Insbesondere faseroptische Raman-Spektrometer verwenden hochwertige Bandpass- und Kerbfilter, um die Rayleigh-Streustrahlung, die den Detektor erreicht, zu minimieren.
Der Detektor – oft ein ladungsgekoppeltes Gerät oder eine Photomultiplier-Röhre – wandelt das optische Signal in ein elektrisches um und erzeugt ein Raman-Spektrum.
Raman-Instrumente mit Fourier-Transformation verwenden eine Dauerstrich-Laserquelle zusammen mit einem Michelson-Interferometer anstelle eines Monochromators, wobei die Strahlung zur Analyse auf eine gekühlte Fotodiode fokussiert wird.

Raman Spectroscopy Instrumentation: Overview

Ein herkömmliches Raman-Spektrophotometer umfasst eine Laserquelle, eine Halterung für Proben, eine Komponente zum Filtern der Wellenlänge und einen Detektor.

Die monochromatische Laserquelle, die typischerweise sichtbare oder nahinfrarote Strahlung verwendet, erzeugt einen hochfokussierten Lichtstrahl. Dieses Licht interagiert mit den Molekülen der Probe und streut einen Teil des Lichts. Flüssige und gasförmige Proben werden normalerweise in gewöhnlichen Glaskapillaren getestet, während Feststoffe als in Kapillaren verpackte Pulver oder als Kaliumbromidpellets analysiert werden können. Das optische Signal wird dann im Detektor, häufig ein Charge-Coupled Device oder ein Photomultiplier, in ein elektrisches Signal umgewandelt, sodass es verarbeitet und als Raman-Spektrum visualisiert werden kann.

In einigen Fällen werden in faseroptischen Raman-Spektrometern hochwertige Bandpass- und Bandsperrfilter verwendet, um die Rayleigh-Streustrahlung zu minimieren. Eine andere Variante, das Fourier-Transform-Raman-Instrument, ersetzt den Monochromator durch ein Michelson-Interferometer und verwendet einen Dauerstrichlaser. Raman spectroscopy uses a laser, a sample holder, a wavelength selector, and a detector to measure scattered light from a sample. The laser is monochromatic, meaning it has one main wavelength. It usually uses visible or near-infrared light to create a tightly focused beam.

That beam hits the sample and some of the light scatters. Liquid and gas samples are often placed in ordinary glass capillaries. Solid samples can be tested as powders packed into capillaries or as potassium bromide pellets.

The scattered light is collected by a separate lens and sent into a monochromator, which separates the light into its different frequencies. The output is then filtered carefully to remove stray laser light and Rayleigh scattering, which can hide the Raman signal. In fiber-optic Raman instruments, high-quality bandpass and notch filters are often used to reduce Rayleigh-scattered light.

Next, the optical signal is changed into an electrical signal by the detector. A charge-coupled device, or CCD, or a photomultiplier tube may be used. The signal can then be processed and displayed as a Raman spectrum.

Some Raman instruments use a different design called Fourier Transform Raman. In this setup, the monochromator is replaced by a Michelson interferometer, and a continuous-wave laser is used. After filtering, the radiation is focused onto a cooled germanium detector for analysis.
