Atomic Spectroscopy: Absorption, Emission, and Fluorescence

Die Atomspektroskopie ist ein analytisches Werkzeug für die qualitative und quantitative Elementaranalyse.
Zu den wichtigsten Typen gehören die Atomabsorptionsspektroskopie (AAS), die Atomemissionsspektroskopie (AES) und die Atomfluoreszenzspektroskopie (AFS).
Alle drei Methoden beginnen mit der Zerstäubung, der kritischen Stufe bei der Zerlegung flüssiger Proben in Gasphasenatome.
Bei der AAS absorbieren heiße gasförmige Proben, die mit einer Reihe elektromagnetischer Strahlung bestrahlt werden, Licht der spezifischen Wellenlängen, die in den Atomen elektronische Übergänge zu Orbitalen mit höherer Energie verursachen.

Die Atomspektroskopie ist ein wichtiges Instrument der Elementaranalyse, sowohl qualitativ als auch quantitativ. Sie kann grob in optische Spektroskopie, Massenspektroskopie und Röntgenspektroskopie unterteilt werden. Zu den optischen spektroskopischen Methoden gehören die Atomabsorptionsspektroskopie (AAS), die Atomemissionsspektroskopie (AES) und die Atomfluoreszenzspektroskopie (AFS). Der erste Schritt bei allen drei Methoden ist die Atomisierung, bei der Feststoffe, Flüssigkeiten oder Proben in Lösung in gasförmige Atome und Ionen umgewandelt werden.

Bei der AAS interagieren die gasförmigen Proben mit elektromagnetischer Strahlung und absorbieren Photonen mit genau den Energien, die Elektronen von Atomen im Grundzustand in ihren angeregten Zustand versetzen. Beispielsweise wird das ungepaarte 3s-Elektron des Na-Atoms bei der Absorption von Strahlung von 285 nm, 330 nm bzw. 590 nm in das 3p-, 4p- oder 5p-Orbital befördert. Die Abnahme des transmittierten Lichts bestimmter Wellenlängen wird vom Detektor gemessen und mit einem Absorptions- oder Transmissionsspektrum visualisiert.

Bei der AES werden die Gasphasenatome im Grundzustand mit Wärme oder elektrischer Entladungsenergie elektronisch angeregt. Diese kurzlebigen, hochenergetischen angeregten Gasphasenatome relaxieren wieder in den Grundzustand und emittieren Photonen, die der Energiedifferenz entsprechen. Die Intensität des emittierten Lichts wird erfasst und in ein elektrisches Signal umgewandelt, das einen Fingerabdruck der Probe liefert. Beispielsweise führen elektronische Übergänge angeregter Na-Atome von den 3p-, 4p- und 5p-Orbitalen zurück zum 3s-Orbital zu Emissionen bei etwa 590 nm, 330 nm bzw. 285 nm. Die emittierte Strahlung wird gemessen und zu einem Spektrum verarbeitet.

Bei der AFS werden die Atome im Grundzustand der Gasphase mit einer charakteristischen Wellenlänge bestrahlt und in den elektronisch angeregten Zustand versetzt. Sofern kein strahlungsloser Übergang stattfindet, relaxieren sich die Atome im angeregten Zustand in den Grundzustand, indem sie bei der genauen Wellenlänge fluoreszieren, die der von ihnen absorbierten Energie entspricht. Der Detektor befindet sich normalerweise im rechten Winkel zum Quellstrahl, sodass ausschließlich Fluoreszenzemissionen erfasst werden.

Im Gegensatz zu Molekülspektren weisen Atomspektren scharfe Linien auf, da verschiedene Rotations- und Schwingungsenergiezustände fehlen, die zu einer Peakverbreiterung in Molekülspektren führen.

Atomic spectroscopy is used to identify and measure elements by their light signals. It supports both qualitative analysis, which tells what is present, and quantitative analysis, which tells how much is present. The optical methods in this group are atomic absorption spectroscopy, atomic emission spectroscopy, and atomic fluorescence spectroscopy.

These methods begin with atomization. In this step, a solid, liquid, or solution sample is changed into gas-phase atoms and ions. Once the sample is in the gas phase, each method uses light or energy in a different way to study the atoms.

In atomic absorption spectroscopy, gas-phase atoms absorb electromagnetic radiation with the exact energy needed to move electrons to higher levels. For example, the 3s electron of sodium can be promoted to the 3p, 4p, or 5p orbital by light of 285 nm, 330 nm, or 590 nm. The detector measures the drop in transmitted light at selected wavelengths and shows it as an absorbance or transmittance spectrum.

In atomic emission spectroscopy, ground-state gas-phase atoms are excited by heat or electrical discharge energy. These excited atoms quickly return to the ground state and release photons that match the energy gap. The emitted light is detected and converted to an electrical signal that gives a fingerprint of the sample. For sodium, transitions from the 3p, 4p, and 5p orbitals back to 3s produce emissions around 590 nm, 330 nm, and 285 nm.

Atomic fluorescence spectroscopy also starts with ground-state gas-phase atoms. They are irradiated with a characteristic wavelength and moved to an excited state. If radiationless transition does not occur, the atoms return to the ground state by fluorescing at the same wavelength they absorbed. The detector is usually placed at a right angle to the source beam so it measures mostly the fluorescence signal.

Atomic spectra are different from molecular spectra because they have sharp lines. Molecules have rotational and vibrational energy states that can broaden peaks, but atoms do not show that same type of broadening. That is why atomic spectra often appear as narrow, distinct signals.
