AAS zerstäubt Proben in der Regel durch Flammen- oder elektrothermische Zerstäubung.
Bei der Flammenzerstäubung wird in der Regel ein Vernebler verwendet, um die Probe kontinuierlich zu aerosolisieren, und eine Sprühkammeranordnung, um sie mit Kraftstoff und Oxidationsmittel zu mischen.
Nur etwa fünf Prozent der Aerosoltröpfchen sind fein genug, um die Flamme zu erreichen, wo sie sich in der primären Verbrennungszone auflösen und blanke Partikel hinterlassen, die in der inneren Flamme zerstäubt werden.
Die gasförmigen Atome, Ionen und Molekülspezies strömen zur Analyse schnell durch den interzonalen Bereich und aus der Flamme heraus.
Da nur sehr wenig von der Probe erfolgreich zerstäubt und detektiert wird, ist die Flammenzerstäubung nicht gut für Proben mit niedrigen Analytkonzentrationen oder begrenzten Volumina.
Auf der anderen Seite wird bei der elektrothermischen Zerstäubung, auch bekannt als Graphitofenzerstäubung, ein Graphitrohr verwendet, um Analyten aus kleinen, diskreten Proben einzufangen und zu konzentrieren, die bei hohen Temperaturen getrocknet, verkohlt und zerstäubt werden.
Alternativ können einige Elemente unter milderen chemischen Bedingungen zuerst in flüchtige Hydridprodukte umgewandelt und dann zerstäubt werden. Darüber hinaus kann bei der Quecksilberbestimmung aufgrund seiner natürlichen Flüchtigkeit eine einzigartige Kaltdampfmethode verwendet werden.

Bei der Atomabsorptionsspektroskopie (AAS) werden die Proben durch Flammenatomisierung oder elektrothermische Atomisierung atomisiert. Bei der Flammenatomisierung wird in der Regel eine  Zerstäuber-Sprühkammer-Anordnung eingesetzt, um die Probe mit einem Brennstoff-Oxidationsmittel-Gemisch zu  kombinieren, wodurch ein feiner Aerosolnebel entsteht, der in einen Brenner gelangt. Normalerweise werden Brennstoff und Oxidationsmittel in einem annähernd stöchiometrischen Verhältnis kombiniert. Für leicht oxidierbare Atome kann jedoch ein brennstoffreiches Gemisch vorteilhafter sein. Nur etwa 5 % der Aerosoltröpfchen erreichen die Flamme, wo sie in der primären Verbrennungszone einer Desolvatisierung unterzogen werden, wodurch bloße Partikel in der inneren Flamme atomisiert zurückbleiben. Die gasförmigen Atome, Ionen und molekularen Spezies durchlaufen schnell den interzonalen Bereich zur Analyse, bevor sie die Flamme wieder verlassen. Flammenzerstäuber haben eine geringe Zerstäubungseffizienz, da große Aerosoltröpfchen die Flamme nicht erreichen und die Probe durch die Verbrennungsgase stark verdünnt wird. Die Effizienz der Flammenzerstäubung kann jedoch durch kontinuierliches Ansaugen der Probe, Optimieren des Brennstoff-Oxidationsmittel-Verhältnisses, Anpassen der Zerstäuberdurchflussraten und Einstellen der Brennerhöhe verbessert werden.

Die Flammenatomisierung ist nicht geeignet für Proben mit niedrigen Analytkonzentrationen oder begrenztem Probenvolumen, da nur ein kleiner Teil der Probe erfolgreich atomisiert und detektiert wird. Im Gegensatz dazu verwendet die elektrothermische Atomisierung, auch als Graphitrohratomisierung bekannt, ein Graphitrohr, um die Analyten einzufangen und zu konzentrieren und eignet sich gut für kleine, diskrete Proben. Bei dieser Technik wird die Probe getrocknet und verkohlt, bevor sie bei hohen Temperaturen atomisiert wird.

Elemente wie As, Se, Sb, Bi, Ge, Sn, Te und Pb können unter milderen Bedingungen atomisiert werden, indem sie chemisch in flüchtige Hydride umgewandelt werden, bevor sie zur Flamme transportiert werden. Zudem kann Quecksilber aufgrund seiner natürlichen Flüchtigkeit mittels einer speziellen Kaltdampfmethode bestimmt werden.

Atomic absorption spectroscopy uses sample atomization to turn a liquid specimen into free atoms for analysis. The two main approaches are flame atomization and electrothermal atomization. Both methods prepare the sample so its atoms can be measured by the instrument.

Flame atomization uses a nebulizer and spray chamber to mix the sample with a fuel-oxidant mixture. This creates a fine aerosol mist that enters the burner. The fuel and oxidant are usually close to a stoichiometric ratio, but a fuel-rich mixture can work better for atoms that oxidize easily.

Only about 5% of the aerosol droplets reach the flame. In the primary combustion zone, these droplets undergo desolvation, which means the solvent is removed. The bare particles are then atomized in the inner flame. The gaseous atoms, ions, and molecular species move through the interzonal region for analysis before leaving the flame.

Flame atomizers have low atomization efficiency. Large droplets do not reach the flame, and combustion gases also dilute the sample. Efficiency can be improved by continuously aspirating the sample, adjusting the fuel-to-oxidant ratio, changing the nebulizer flow rate, and setting the burner height. Flame atomization is not a good choice for samples with very low analyte levels or small sample volumes.

Electrothermal atomization, also called graphite furnace atomization, uses a graphite tube to collect and concentrate the analytes. It works well for small, discrete samples. The sample is first dried and charred, then atomized at very high temperatures.

Some elements, including As, Se, Sb, Bi, Ge, Sn, Te, and Pb, can be handled under milder conditions by forming volatile hydrides before they are carried to the flame. Mercury can also be measured with a cold-vapor method because it is naturally volatile.
