Atomic Emission Spectroscopy: Lab

ICP ist aufgrund seiner hohen Stabilität, seines geringen Rauschens und seiner Interferenzfreiheit unter genauen experimentellen Bedingungen eine bevorzugte Quelle für AES.
Es analysiert üblicherweise Lösungsproben. Zusätzlich werden feste Proben direkt mittels elektrothermischer Verdampfung oder Laser- und Funkenablation analysiert.
Mit der Plasmaemissionsspektrometrie können etwa 60 Elemente aus dem Periodensystem qualitativ identifiziert und quantifiziert werden.
Elemente wie Bor, Kohlenstoff, Stickstoff, Phosphor und Schwefel erfordern jedoch ein Vakuumspektrometer, da ihre Emissionslinien unter 180 nm fallen, was zu Interferenzen durch atmosphärische Komponenten führt.
Die Alkalimetalldetektion stellt eine Herausforderung dar, da die Betriebsbedingungen ungeeignet sind und die Nahinfrarot-Emissionsleitungen eingehalten werden.
In der Plasmaemissionsspektroskopie sind Kalibrierkurven lineare Diagramme elektrischer Signale, die proportional zur Linienintensität gegen die Analytkonzentration sind.
Für die quantitative Analyse werden üblicherweise externe oder interne Standards mit ähnlichen Emissionslinien und chemischen Interferenzen wie der Analyt verwendet.
Interne Standards werden jedoch bevorzugt, wenn Faktoren wie die Temperatur der Anregungsquelle und die Zerstäubungseffizienz die Emissionsintensität unkontrollierbar beeinflussen.

Insbesondere das induktiv gekoppelte Plasma (ICP) liefert aufgrund seiner hohen Stabilität, des geringen Rauschens, des geringen Hintergrunds und der minimalen Interferenzen bei optimalen Versuchsbedingungen überlegene quantitative Analysedaten. Neuere luftbetriebene Mikrowellen-Plasmafackeln erweisen sich jedoch als vielversprechende Alternativen, die kostengünstiger sein könnten als herkömmliche ICP-Quellen. AES wird hauptsächlich zur Analyse von flüssigen Proben verwendet. Plasmaemission ermöglicht jedoch auch die direkte Analyse von festen Proben, was durch verschiedene Verfahren wie elektrothermische Verdampfung, Laser- und Funkenablation und Glimmentladung erreicht werden kann.

Theoretisch können alle metallischen Elemente durch Plasma-Emissionsspektrometrie bestimmt werden. Die Effizienz dieser Methode für Alkalimetalle ist allerdings aufgrund der anspruchsvollen Betriebsbedingungen und der Lage ihrer markanten Spektrallinien im nahen Infrarotbereich begrenzt. Dies kann in vielen Plasmaspektrometern zu Problemen bei der Detektion führen, da sie hauptsächlich für ultraviolette Strahlung ausgelegt sind. Daher ist die Plasma-Emissionsspektroskopie im Allgemeinen auf die Bestimmung von etwa 60 Elementen beschränkt. Die meisten Elemente haben mehrere markante Emissionslinien, die sich zur Identifizierung und Quantifizierung eignen. Es ist also normalerweise möglich, eine geeignete Linie zur Bestimmung eines beliebigen Elements zu identifizieren. Die Wahl der Linie hängt von der Einschätzung ab, welche anderen Elemente in der Probe vorhanden sein könnten. Bei der Linienauswahl für das untersuchte Element muss die mögliche Überlappung der Linien anderer Elemente vermieden werden. 

Plasmaquellen liefern oft lineare Kalibrierungskurven, aber Abweichungen von der Linearität können aufgrund von Faktoren wie Selbstabsorption, fehlerhaften Hintergrundkorrekturen, Ionisierung und nichtlinearen Reaktionen der Detektionssysteme auftreten. Wenn möglich, werden quantitative Analysen am besten mit externen Standards durchgeführt. 

Die Emissionsintensität kann jedoch von vielen Parametern erheblich beeinflusst werden, darunter die Temperatur der Anregungsquelle und die Zerstäubungseffizienz. In Fällen, in denen diese Parameter schwer zu kontrollieren sind, können interne Standards verwendet werden.

Atomic emission spectroscopy uses plasma sources to identify elements by their light. The method creates abundant spectra with characteristic emission lines. Inductively coupled plasma, or ICP, is especially useful because it gives strong quantitative data. It works well under stable conditions, with low noise, low background, and few interferences.

Newer air-operated microwave sources are also being studied. These sources may be more cost-effective than conventional ICP systems. Atomic emission spectroscopy is mainly used for liquid samples, but plasma emission can also be used for solid samples. Direct solid analysis can be done with electrothermal vaporization, laser and spark ablation, or glow-discharge vaporization.

In theory, plasma emission spectrometry can determine all metallic elements. In practice, its use for alkali metals is limited. Their strong spectral lines are in the near-infrared region, which can be hard to detect in many instruments made mainly for ultraviolet light. Because of this, plasma emission spectroscopy is usually used for about 60 elements.

Most elements have several strong lines that can help with both identification and measurement. A good line is usually chosen by checking overlap with lines from other elements in the sample. Plasma sources often give linear calibration curves, but some curves can deviate from linearity. This can happen because of self-absorption, background correction errors, ionization, or detector response problems.

When possible, quantitative analysis is best done with external standards. These are known samples used to compare emission intensity. Emission intensity can also change with source temperature and atomization efficiency, which is how well a sample turns into atoms. When source conditions are hard to keep constant, internal standards can help improve the measurement.
