Subviral Agents

Subvirale Erreger - Viroide, Prionen und Satelliten - ähneln Viren, haben aber keine viralen Komponenten. Viroide bestehen aus einer einzelsträngigen RNA, haben kein Kapsid und infizieren nur Pflanzen. Sie verlassen sich auf die RNA-Polymerase des Wirts, um sich zu replizieren und sich selbst zu spalten, um RNA-Einheiten freizusetzen. Diese RNAs kodieren nicht für Proteine, sondern bilden stabile Sekundärstrukturen, die als kleine interferierende RNAs fungieren, die Pflanzengene zum Schweigen bringen. Prionen sind Infektionserreger, die vollständig aus Proteinen bestehen und denen Nukleinsäuren fehlen. PrPC ist ein normales Protein bei Tieren. Wenn infektiöses PrPSc in die Zellen eindringt, faltet es PrPC falsch in PrPSc und bildet schädliche Amyloid-Aggregate, die die Neuronen schädigen. Dies führt zu transmissiblen spongiformen Enzephalopathien, die durch Hirndegeneration und Vakuolenbildung gekennzeichnet sind. Prionenkrankheiten können vererbt oder durch infiziertes Fleisch oder chirurgische Instrumente erworben werden. Beispiele hierfür sind die Traberkrankheit bei Schafen, der Rinderwahnsinn bei Rindern und die Creutzfeldt-Jakob-Krankheit beim Menschen. Satelliten sind DNA oder RNA, die in einem Kapsid eingeschlossen sind. Für die Replikation sind sie auf Hilfsviren angewiesen.

Viroide sind die einfachsten subviralen Agenzien und bestehen aus zirkul&#228;ren, einzelstr&#228;ngigen RNA-Molek&#252;len ohne Proteinh&#252;lle. Sie infizieren ausschlie&#223;lich Pflanzen und sind vollst&#228;ndig auf die zellul&#228;re Maschinerie des Wirts f&#252;r ihre Replikation angewiesen. Viroide werden durch die RNA-Polymerase des Wirts &#252;ber einen Rolling-Circle-Mechanismus repliziert, wobei ihre Spaltung in funktionelle Einheiten durch ribozymatische Aktivit&#228;t erfolgt. Pathogene Viroide umfassen das Das Kartoffelspindelknollen-Viroid (PSTVd), das Kartoffel- und Tomatenpflanzen bef&#228;llt und zu Wachstumshemmung und Ertragsminderung f&#252;hrt.
Prionen sind einzigartige infekti&#246;se Agenzien, die ausschlie&#223;lich aus fehlgefalteten Proteinen bestehen und keine Nukleins&#228;uren enthalten. Das pathogene Prionenprotein, PrP^Sc, ist eine fehlgefaltete Form des normalen zellul&#228;ren Proteins PrP^C. Wenn PrP^Sc mit PrP^C interagiert, induziert es eine Konformations&#228;nderung, die das normale Protein zur Fehlfaltung bringt, welches dann zu Amyloidfibrillen aggregiert. Diese Aggregate akkumulieren im Nervengewebe und verursachen neurodegenerative Erkrankungen, die als transmissible spongiforme Enzephalopathien (TSEs) bekannt sind. TSEs zeichnen sich durch fortschreitende Gehirndegeneration, Neuronenverlust und die Bildung von Vakuolen im Gehirngewebe aus. Prionenerkrankungen k&#246;nnen vererbt werden, spontan auftreten oder durch Kontakt mit kontaminierten Lebensmitteln, chirurgischen Instrumenten oder Transplantaten &#252;bertragen werden. Beispiele sind Scrapie bei Schafen, die bovine spongiforme Enzephalopathie (BSE), auch &#8222;Rinderwahnsinn&#8220;, bei Rindern und die Creutzfeldt&#8211;Jakob-Krankheit (CJK) beim Menschen.
Satelliten sind subvirale Agenzien, die entweder RNA oder DNA enthalten, die in ein Kapsid eingeschlossen ist. Allerdings fehlt ihnen die genetische Information f&#252;r eine unabh&#228;ngige Replikation, weshalb sie auf eine Koinfektion mit einem Helfer-Virus angewiesen sind. Satellitenviren ben&#246;tigen dieses Helfer-Virus zur Replikation und modifizieren h&#228;ufig die durch das Helfervirus verursachten Krankheitssymptome. Das Hepatitis-D-Virus (HDV) ist ein Satelliten-RNA-Virus, das auf das Hepatitis-B-Virus (HBV) f&#252;r seine Replikation und Verpackung angewiesen ist und bei Koinfektion die Lebererkrankung versch&#228;rft.

viroids, prions, and satellite viruses

Viroids, prions, and satellites are subviral agents that can cause infection even though they are not complete viruses. They resemble viruses in spread and disease, but each lacks one or more key viral parts, such as a capsid or the machinery needed for replication.

Viroids are the simplest of these agents. They are circular, single-stranded RNA molecules with no protein coat. Viroids infect plants only and depend on host cell machinery to copy themselves. Host RNA polymerase replicates them through a rolling-circle mechanism, and ribozyme activity cuts the RNA into functional units. Viroids do not make proteins. Instead, their folded RNA can act as small interfering RNAs, or siRNAs, that disrupt host gene expression and can cause disease. One example is Potato spindle tuber viroid, or PSTVd, which infects potato and tomato plants and can lead to stunted growth and lower yield.

Prions are different because they are made only of misfolded protein and contain no nucleic acids. The harmful form, PrPSc, is an altered version of the normal cellular protein PrPC. When PrPSc meets PrPC, it can trigger the normal protein to misfold and clump together into amyloid fibrils. These aggregates build up in nerve tissue and cause transmissible spongiform encephalopathies, or TSEs. TSEs involve progressive brain damage, loss of neurons, and vacuoles in brain tissue. Prion disease may be inherited, may appear on its own, or may be acquired through contaminated food, surgical instruments, or transplants. Examples include scrapie in sheep, bovine spongiform encephalopathy, or mad cow disease, in cattle, and Creutzfeldt-Jakob disease in humans.

Satellites are subviral agents that contain RNA or DNA inside a capsid, but they cannot replicate on their own. They need co-infection with a helper virus to copy their genetic material. Satellite viruses depend on the helper virus for replication and packaging, and they often change the symptoms caused by the helper virus. Hepatitis D virus, or HDV, is a satellite RNA virus that relies on Hepatitis B virus, or HBV, and can make liver disease worse in people co-infected with both viruses.
