Nano Bead Preparation

Nano bead preparation is the controlled synthesis and surface modification of nanoscale beads, producing particles with defined size, composition, and chemical functionality for biochemical research. In common nanoprecipitation or emulsion-based approaches, rapid mixing creates supersaturation or dispersed droplets that drive particle formation, while surfactants or stabilizing ligands limit aggregation and help control bead size; washing and purification remove unbound materials. Functionalized nanobeads can capture proteins, nucleic acids, or small molecules through affinity interactions, enabling magnetic separation, biosensing, sample enrichment, and targeted delivery studies. Reproducible preparation improves assay consistency and supports the development of diagnostic and analytical tools.

We present a method for achieving sub-nanometer resolution images with amplitude-modulation (tapping mode) atomic force microscopy in liquid. The method is demonstrated on commercial atomic force microscopes. We explain the rationale behind our choices of parameters and suggest strategies for resolution optimization.

A protocol for the preparation of poly(pentafluorophenyl acrylate) (poly(PFPA)) grafted silica beads is presented. The poly(PFPA) functionalized surface is then immobilized with antibodies and used successfully for the protein separation through immunoprecipitation.

The on-bead method for labeling antibodies with small molecules enables labeling of a small amount of antibodies directly from cell media. This method is compatible with amine and thiol chemistry, and can handle multiple samples in parallel, manually or using automated platforms.

Mice are generally resistant to infections by Mycobacterium abscessus, which complicates the discovery and development of much-needed antibiotics against pulmonary infections. Here, we describe a method of inoculum preparation and intratracheal infection that was shown to deliver sustained infection in immunocompetent mice.
