Wharton's Jelly Mscs

Wharton's jelly mesenchymal stromal cells (WJ-MSCs) are multipotent, nonhematopoietic cells isolated from the gelatinous connective tissue surrounding umbilical vessels, making them valuable for regenerative research and biochemistry. In culture, they adhere to plastic, expand under defined conditions, and respond to biochemical cues through signaling pathways that regulate self-renewal, differentiation, extracellular-matrix production, and secretion of paracrine factors. Researchers use WJ-MSCs to study cell metabolism, protein expression, immunomodulatory signaling, and tissue repair, while their umbilical-cord origin provides an accessible source for investigating stem-cell behavior and developing cell-based approaches for injury, inflammation, and regenerative medicine.

Cannulation of the Mouse Submandibular Salivary Gland via the Wharton's Duct

A protocol for the cannulation of the mouse submandibular salivary gland via the Wharton's duct is described. For this experiment, the trypan blue solution is used as a dyer to demonstrate how this technique effectively delivers infusions into the targeted gland, and to suggest the reliability of this new approach as a potential clinical drug/cell therapy for the regeneration of salivary glands.

Isolation and Enrichment of Rat Mesenchymal Stem Cells (MSCs) and Separation of Single-colony Derived MSCs

Rat MSCs were isolated from femurs and tibias and then enriched by magnetic cell sorting. Sorted cells were confirmed for the expression of surface markers by flow cytometry. These cells were also cultured at clonal density to form single colonies and then these colonies were separated by cloning cylinders.

The immunomodulatory properties of human mesenchymal stem cells (MSC) appear increasingly relevant for clinical application. Using a co-culture system of MSCs and peripheral blood leukocytes pre-stained with the fluorescent dye carboxyfluorescein succinimidyl ester (CFSE), we describe the in vitro assessment of MSC immunomodulation on effector leukocyte proliferation and specific subpopulations.
