Biotin Attachment

Biotin attachment is the covalent linking of biotin, a small vitamin-derived molecule, to proteins, nucleic acids, surfaces, or other biomolecules to create a versatile molecular handle. The attached biotin binds avidin or streptavidin with exceptionally high affinity, enabling selective capture, immobilization, or detection; chemistries such as NHS ester or maleimide coupling can target amine or thiol groups under controlled conditions. In bioengineering, this interaction supports protein purification, biosensor construction, molecular imaging, assay development, and the organization of biomolecules on engineered surfaces. Its strong yet selective binding helps connect biological components with analytical and material platforms.

RNA/protein complexes purified using botin-streptavidin strategy are eluted to solution under denaturing conditions in a form unsuitable for further purification and functional analysis. Here, we describe a modification of this strategy that utilizes a photo-cleavable linker in RNA and a gentle UV-elution step, yielding native and fully functional RNA/protein complexes.
