Dna Nanoparticle Transfection

DNA nanoparticle transfection is a nonviral gene-delivery technique that introduces nucleic acid cargo into cells for controlled gene expression, making it useful for bioengineering and biomedical research. In this approach, plasmid DNA or other DNA constructs are assembled with nanoparticle materials that protect them from degradation and promote cellular uptake, often through endocytosis; intracellular processing can then release the DNA for transport to the nucleus and transcription. DNA nanoparticle transfection supports studies of gene function, engineered cells, tissue regeneration, and potential therapeutic delivery. By tuning particle size, surface chemistry, cargo loading, and exposure conditions, researchers can influence transfection efficiency, cell compatibility, and the duration of gene expression, advancing safer alternatives to viral vectors.

Type B gelatin-based engineered nanovectors system (GENS) was developed for systemic gene delivery and transfection in the treatment of pancreatic cancer. By modification with epidermal growth factor receptor (EGFR) specific peptide on the surface of nanparticles, they could target on EGFR receptor and release plasmid under reducing environment, such as high intracellular glutathione concentrations.
