Dynamic Confocal Slit

Dynamic confocal slit is an adjustable optical aperture used in confocal microscopy to control how light reaches the detector and improve separation of in-focus image information from background signal. During image acquisition, the slit width can vary with imaging conditions, balancing illumination, detection efficiency, spatial resolution, and optical sectioning while limiting out-of-focus light. In bioengineering, this approach supports fluorescence imaging of cells, tissues, and engineered biological systems, where adjustable detection can help capture three-dimensional structure and dynamic processes with improved contrast and reduced unnecessary light exposure. It is especially useful when imaging conditions change across a sample or experiment.

Fabrication and Visualization of Capillary Bridges in Slit Pore Geometry

A procedure for creating and imaging capillary bridges in slit-pore geometry is presented. The creation of capillary bridges relies on the formation of pillars to provide a directional physical and chemical heterogeneity to pin the fluid. Capillary bridges are formed and manipulated using microstages and visualized using a CCD camera.

This manuscript describes the design and operation of a microtensiometer/confocal microscope to do simultaneous measurements of interfacial tension and surface dilatational rheology while visualizing the interfacial morphology. This provides the real-time construction of structure-property relationships of interfaces important in technology and physiology.

Microglia activation and microgliosis are key responses to chronic neurodegeneration. Here, we present methods for in vivo, long-term visualization of retinal CX3CR1-GFP+ microglial cells by confocal ophthalmoscopy, and for threshold and morphometric analyses to identify and quantify their activation. We monitor microglial changes during early stages of age-related glaucoma.
