Parallel Artificial Membrane

Parallel artificial membrane is an in vitro technique that models passive transport across biological barriers by measuring how compounds cross a synthetic lipid membrane. In a typical assay, a lipid solution forms a membrane over a porous support separating donor and acceptor compartments; compounds diffuse according to concentration gradients, and sampling over time yields an apparent permeability value. In bioengineering, the method provides a rapid, reproducible screen for membrane permeability before more complex cell-based or animal studies. It supports drug development, formulation design, and comparison of how molecular properties such as lipophilicity and ionization influence transport.

The presented protocol describes the analysis of membrane protein mediated transport on the single transporter level using pore-spanning solvent-free lipid bilayers. This is achieved by the creation of bulk produced nanopore array chips, combined with highly parallel data acquisition and analysis, enabling the future establishment of membrane protein effector screenings.
