Size Heterogeneity Analysis

Size heterogeneity analysis is a biological measurement approach that quantifies how much individual cells, particles, vesicles, or biomolecules vary in physical size within a sample. Under controlled conditions, analytical measurements capture size-dependent signals, such as elution behavior, diffusion, imaging dimensions, or light scattering, and convert them into a size distribution rather than a single average value. This analysis can reveal subpopulations, aggregation, fragmentation, or sample inconsistency that bulk measurements may obscure. In biological research and bioprocessing, it supports characterization, quality control, experimental reproducibility, and evaluation of how biological conditions or treatments alter sample composition.

Bacterial vesicles play important roles in pathogenesis and have promising biotechnological applications. The heterogeneity of vesicles complicates analysis and use; therefore, a simple, reproducible method to separate varying sizes of vesicles is necessary. Here, we demonstrate the use of size exclusion chromatography to separate heterogeneous vesicles produced by Aggregatibacter actinomycetemcomitans.

Here we describe a method to quantify molecular heterogeneity in histological sections of tumor material using quantitative immunofluorescence, image analysis, and a statistical measure of heterogeneity. The method is intended for use in clinical biomarker development and analysis.
