Densitometry

Densitometry is a quantitative technique that measures the density of a material, sample, or image to estimate its composition or concentration. In biological research, it commonly analyzes how much light a sample absorbs or transmits, converting optical intensity into a numerical value through calibration against standards; related methods can assess mineral density using X-ray attenuation. Densitometry supports protein and nucleic acid analysis, gel-band quantification, cell and tissue imaging, and bone-density assessment. By transforming visual or physical differences into reproducible measurements, it enables comparisons among experimental groups and strengthens the interpretation of biological assays.

Western Blotting is used to identify the presence of specific proteins in electrophoretically separated samples. Following separation by a technique known as sodium dodecyl sulfate polyacrylamide gel electrophoresis, or SDS-PAGE, western transfer is used to move proteins from a polyacrylamide gel onto a piece of membrane which traps the proteins in their respective locations. Testing protein-protein interaction is indispensable for dissection of protein functionality. Here, we introduce an in vitro protein-protein binding assay to probe a membrane-immobilized protein with a soluble protein. This assay provides a reliable method to test interaction between an insoluble protein and a protein in solution.

Here we present a protocol to describe the localization of angiotensin II Type 1 receptors in the rat brain by quantitative, densitometric, in vitro receptor autoradiography using an iodine-125 labeled analog of angiotensin II.
