DNA polymerase is an enzyme that synthesizes DNA, making it essential for genome replication, repair, and inheritance. During replication, it reads a template strand and adds complementary deoxynucleotides to the growing strand in the 5′-to-3′ direction, using base pairing and often proofreading to correct mismatches. In biology, DNA polymerases support accurate chromosome duplication and help maintain genetic stability; their activity also underlies laboratory methods such as polymerase chain reaction (PCR), DNA sequencing, and molecular cloning. Studying their structure and function clarifies how mutations arise and enables genetic analysis and biotechnology.
