Fdu Inhibition

FdU inhibition is the suppression of cellular DNA synthesis by 5-fluoro-2′-deoxyuridine (FdU), a fluorinated nucleoside analog used to study cell proliferation and nucleotide metabolism. Inside cells, FdU is converted to 5-fluoro-2′-deoxyuridine monophosphate (FdUMP), which forms a stable inhibitory complex with thymidylate synthase and reduced folate. This blocks conversion of dUMP to dTMP, depleting the thymidine required for DNA replication. In biology, FdU inhibition helps researchers arrest cell-cycle progression, assess DNA damage and replication stress, and evaluate cellular responses to antimetabolite treatment, with applications in cancer biology and drug-sensitivity studies.

Habituation and prepulse inhibition of startle are operational measures of sensory gating. Sensory gating is disrupted in schizophrenia, and some other mental disorders and neurodegenerative diseases. We here describe a standard protocol to assess short-term and long-term habituation as well as prepulse inhibition of acoustic startle responses in rats and mice.
