Mesenchymal Stem Cells

Mesenchymal stem cells (MSCs) are multipotent stromal cells that can self-renew and generate several connective-tissue cell types, making them important subjects in biology and regenerative research. Their behavior depends on signals from the surrounding environment: culture conditions and molecular cues can direct differentiation into osteoblasts, chondrocytes, or adipocytes, while secreted factors can influence inflammation, cell survival, and tissue repair. Researchers use MSCs to study cell differentiation, tissue development, disease mechanisms, and therapeutic strategies, including applications in bone and cartilage repair. Understanding how MSCs respond to biochemical and mechanical signals may improve cell-based treatments and tissue-engineering approaches.

An Assay to Study the Regulation of Macrophage Phagocytosis by Mesenchymal Stem Cells

This video demonstrates an assay to study mesenchymal stem cell (MSC) regulation of macrophage phagocytosis in a co-culture. Upon co-culturing MSC with macrophages, non-opsonized zymosan particles conjugated to a pH-sensitive fluorophore are added to be phagocytosed by the macrophages. With the increase in phagocytosis, the fluorescence intensity rises, allowing the determination of MSC-mediated regulation of phagocytic activity.

Here, we present a method to efficiently harness the cardiac differentiation potential of young sources of human mesenchymal stem cells in order to generate functional, contracting, cardiomyocyte-like cells in vitro.

The immunomodulatory properties of human mesenchymal stem cells (MSC) appear increasingly relevant for clinical application. Using a co-culture system of MSCs and peripheral blood leukocytes pre-stained with the fluorescent dye carboxyfluorescein succinimidyl ester (CFSE), we describe the in vitro assessment of MSC immunomodulation on effector leukocyte proliferation and specific subpopulations.
