Pit And Fissure Sealing

Pit and fissure sealing is a preventive dental procedure that covers the narrow grooves on tooth surfaces, where plaque can accumulate and initiate decay. A resin-based or glass ionomer material flows into these irregularities and hardens or sets, forming a barrier that limits bacterial retention and contact between fermentable food substances and enamel. By reducing the conditions that promote acid production and enamel demineralization, sealants help prevent occlusal caries, particularly in newly erupted permanent molars with deep fissures. In biology and oral health research, the procedure illustrates how modifying a local surface environment can interrupt microbial disease processes and preserve tooth structure.

Key procedures to optimize the sealing process and achieve real-time monitoring of the metal-to-glass seal (MTGS) structure are described in detail. The embedded fiber Bragg grating (FBG) sensor is designed to achieve online monitoring of temperature and high-level residual stress in the MTGS with simultaneous environmental pressure monitoring.

In this video, we describe an in vitro osteoclastic resorption assay on calcium phosphate-coated cell culture plates cultured with human peripheral blood mononuclear cell-derived osteoclasts. The osteoclasts form a characteristic actin ring on the calcium phosphate-coated matrix of the cell culture plate and degrade the matrix within the actin ring region to release calcium and phosphate ions, resulting in the formation of distinct resorption pits within the degraded matrix.

Intended for labs setting-up chip models and small-scale chip platform development. We demonstrate the protocol using plastic and native tissue membranes. This procedure only requires PDMS, toluene, mold, membrane, and a vacuum chamber.
