Gradient Elution

Gradient elution is a chromatographic technique in which the mobile-phase composition changes during a separation, helping resolve compounds with a wide range of affinities for the stationary phase. In liquid chromatography, the solvent strength is typically increased over time, causing strongly retained molecules to migrate through the column and elute after less strongly retained compounds; adjusting the gradient can improve peak shape, resolution, and analysis time. Chemists use gradient elution in high-performance liquid chromatography and related methods to analyze complex mixtures, including samples containing compounds that would be difficult to separate under a single, constant solvent composition. Its controlled solvent changes support efficient, reproducible chemical analysis.
