OPLS-DA, or orthogonal partial least squares discriminant analysis, is a supervised multivariate method that models differences between predefined classes in complex chemical data. It decomposes measured variables into predictive components, which capture variation correlated with class membership, and orthogonal components, which describe systematic variation unrelated to the classification outcome. By separating these sources of variation, OPLS-DA can simplify interpretation of spectroscopy, chromatography, and metabolomics datasets, helping identify chemical features associated with different samples or conditions. Model performance should be assessed with appropriate validation because excessive complexity can produce overfitting and misleading class separation.
